Objective: By investigating the difference in telomere length and the frequency distribution of telomere length-related gene SNPs between the normal population of Hainan Li nationality and the COPD,to get the telomere length-related gene polymorphism and telomere length were found to be associated with the susceptibility of COPD in Hainan Li.The relationship provides more theoretical basis for the pathogenesis of COPD.Methods: A case-control study to collect 279 patients with COPD and 290 normal controls in Hainan Li nationality from 2017 to 2018.After the whole genome DNA was extracted,the SNP typing of 25 loci was carried out by Sequenom MassARRAY technology platform.Chi-square analysis and various genetic model typing results were analyzed statistically.Haploview software calculated the degree of linkage among gene loci and the construction of haplotypes.To determine the relationship between polymorphisms of telomere length-related genes and the risk of COPD in Li.The telomere length of peripheral blood cells was measured by qPCR in patients of COPD and controls.We used the unconditional logistic regression analysis to calculate odds ratio(OR)and 95% confidence interval(95%CI)to analyze the relationship between telomere length and risk of COPD in Li nationality.At the same time,to analyze the relationship between ZNF208 gene polymorphism and telomere length.Results: In the genetic model,the“C/C”genotype of rs4809324 locus on the RTEL1 gene were reduced the risk of COPD in Li base on co-dominant model and recessive model.The “C/C” genotype of rs10069690 locus on the TERT gene were increased the risk of COPD base on Overdominant model.In the Log-additive model,rs2853677,rs2853676 on the TERT gene can increase the risk of COPD in the Li ethnic group.Haploview software analysis showed that there was a strong linkage between the SNPs loci of TERT gene,OBFC1 gene,RTEL1 gene and ZNF208 gene.By measuring the relationship between telomere length and telomere length and gene polymorphism in patients with COPD and controls,we found that the telomere length of Li COPD patients was significantly shorter than that of normal controls.The subjects carrying the G allele of ZNF208 rs2188972 have longer telomere length.Conclusion: The rs4809324 locus on the RTEL1 gene in telomere length-related genes can reduce the risk of COPD in Hainan Li population.The rs10069690,rs2853677 and rs2853676 on the TERT gene can increased the risk of COPD in the Li population of Hainan.Compared with the normal control group,the Li nationality COPD patients have shorter telomere length,and the shorter the telomere length,the higher the risk of COPD.The rs2188972 locus polymorphism in the ZNF208 gene affects telomere length,and the G allele carried by rs2188972 has a longer telomere length.
