Objective:To explore the anti-steatosis effect of Anethole Trithione on non-alcoholic fatty liver disease rats induced by high-fat diet and the possible mechanism.Method:After 1 week of adaptive feeding,50 healthy male rats were randomly divided into control group?CG,n=9?and high-fat group?n=41?,respectively fed with normal chow and high-fat diet.After 11 weeks,the high-fat group was divided into five groups:the empty model group?n=8?,the vitamin E treated group(VE200,200 mg·kg-1·d-1,n=8),the high-dose of Anethole Trithione group(AT20,20mg·kg-1·d-1,n=8),the medium-dose of Anethole Trithione group(AT10,10mg·kg-1·d-1,n=8),the low-dose of Anethole Trithione group(AT5,5mg·kg-1·d-1,n=8).The AT20,AT10,AT5 groups were given Anethole Trithione for 8 weeks,and the VE200 group was given vitamin E for 8 weeks,the control group and the empty model group were given 2ml normal saline per day for 8 weeks.After 19 weeks,the body weight of all the rats were measured first,and all of them were anesthetized intraperitoneally.Blood samples were collected from renal arteries.The livers,perirenal fat and epididymis fat were taken out,and their weight were measured too.The liver indexes and visceral fat ratios were calculated.The serum ALT,AST,TG,TC level was detected,and the liver histology was tested by HE stain.The liver TNF-?level was detected by immunohistochemical technique,the liver FAS and PPAR-?protein level was detected by western blot?WB?.Results:?1?Non-alcoholic fatty liver disease rats were induced by high-fat diet for 10 weeks successfully.?2?After 19 weeks of feeding,the rats in high-fat groups showed obvious visceral obesity.Compared with the CG group,the body weight,liver weight,visceral fat weight,liver indexes,visceral fat content and the serum ALT,AST,TG,TC level,TNF-?protein level,FAS protein level significantly increased in high-fat groups?P<0.05?.However,the PPAR-?protein level in high-fat groups groups were lower than those of CG group?P<0.05?.Compared with the empty model group,the body weight,liver weight,visceral fat weight,visceral fat content and the serum ALT,AST,TG,TC level,TNF-?protein level,FAS protein level significantly decreased in the four treated groups?P<0.05?,except for the AT5 group,the liver indexes were decreased in other treated groups compared with the empty model group?P<0.05?.And,except for the AT5 group,the PPAR-? protein level in other treated groups were higher than that of empty model group?P<0.05?.The AT20 group had a most obvious decrease in the body weight,liver weight,visceral fat weight,liver indexes,visceral fat content,serum TG,TC level,FAS protein level than other treated groups?P<0.05?.Except for the AT5 group,the PPAR-?protein level has no statistical significance in other treated gtoups?P>0.05?.?3?The liver HE staining was normal in the CG group,and significant steatosis was observed in the empty model group,treated groups improved in different degrees.Conclusion:?1?Anethole Trithione has an anti-steatosis effect on NAFLD rats.?2?The possible mechanism of Anethole Trithione in anti-steatosis effect of NAFLD rats is to reduce the expression of liver TNF-?,FAS protein and increase the expression of PPAR-?protein.
