Objective: To investigate the effects of SIRT3 protein expression and breast cancer cell proliferation by regulating the expression level of NRF2.Methods: Group:The MCF-7 cell of breast cancer which purchased from Shanghai sangon company and cryopreserved in our laboratory are selected.Through cell transfection technique,a cell line with high expression of NRF2 gene was established as group 1,a cell line with low expression of NRF2 gene was group 2,a cell line with high expression of both NRF2 and SIRT3 gene was group 3,and establish a control group.Test:1.Fluorescence quantitative PCR was used to detect the expression of NRF2 in each group.2.Western blot was used to detect the expression of SIRT3 protein in each group.3.MTT assay was used to detect the proliferation of breast cancer cell lines in each group.Result:1.The relative expression of NRF2 mRNA in group 1 was(55.64±1.34),(0.52±0.08)in group 2,and(53.52±0.65)in group3,compared with negative control group(1.43±0.25),P<0.05,the difference was statistically significant;the expression of NRF2 mRNA ingroup 1 and group 3 was higher than that in negative control group,and the expression in group 2 was lower than that in negative control group.2.The relative expression of SIRT3 protein in group 1 was(0.225±0.018),(0.748±0.040)in group 2,and(0.473±0.056)in group 3,compared with negative control group(0.296±0.022),P<0.05,the difference was statistically significant.The expression of SIRT3 protein in group 1 was lower than that in negative control group,and the expression of group 2and group 3 was higher than that in negative control group.3.MTT detect the cell proliferation in each group:The OD values of group 1 at 24 h,48h and 72 h were(0.542±0.032)(0.896±0.008)(1.390± 0.042).The OD values of group2 were(0.327±0.018)(0.460±0.0008)(0.634±0.030),compared with the negative control group(0.369±0.027)(0.570±0.009)(0.801± 0.015),P<0.05,the difference was statistically significant.4.The OD values of group 3 at 24 h,48h,and 72 h were(0.425±0.034)(0.706±0.018)(1.042±0.041),compared with the negative control group,P<0.05,the difference was statistically significant;compared with group 1,P <0.05,the difference was statistically significant.CONCLUSION:1.Regulation of NRF2 gene expression can cause SIRT3 gene changes,considering the negative correlation between them.2.NRF2 may affect the proliferation of breast cancer cells by regulating the expression of SIRT3.
