Objective: To investigate the expression and biological function of circ EPSTI1 in human gastric cancer tissues and cells and its clinical significance in gastric cancer.Materials and methods:q RT-PCR was used to detect the expression of circ EPSTI1 in gastric cancer tissues and the normal stomach mucosa,as well as in different gastric cancer cells and normal gastric epithelial cells.The relationship between circ EPSTI1 and the different pathological parameters of gastric cancer patients was statistically analyzed.Transfection of si-circ EPSTI1 knockdown the expression of circ EPSTI1 in human gastric cancer SGC-7901 and MGC-803 cells.The effects of si-circ EPSTI1 on the biological behavior of human gastric cancer SGC-7901 and MGC-803 cells were detected by MTT assay,cell scratch assay,Transwell assay,Annexin V-FITC and PI staining,respectively.Results: q RT-PCR showed that the expression level of circ EPSTI1 in gastric cancer tissues was higher than that in the normal stomach mucosa(P<0.01).Correlation analysis showed that circ EPSTI1 is closely correlated with histological grade,TNM stage,and lymph node metastasis in patients with gastric cancer.The expression of circ EPSTI1 was up-regulated in MGC-803,SGC-7901 and AGS cells compared to GES-1 cells(P<0.01).MTT assay showed that si-circ EPSTI1 could inhibit the proliferation of human gastric cancer SGC-7901 and MGC-803 cells,and the difference was statistically significant(P<0.05).The results of scratch test showed that si-circ EPSTI1 could inhibit the migration of human gastric cancer SGC-7901 and MGC-803 cells,and the difference was statistically significant(P<0.05).Transwell invasion assay showed that si-circ EPSTI1 could inhibit the invasion of human gastric cancer SGC-7901 and MGC-803 cells,and the difference was statistically significant(P<0.01).The results of Annexin V-FITC and PI staining showed that si-circ EPSTI1 induced apoptosis of human gastric cancer SGC-7901 and MGC-803 cells,and the difference was statistically significant(P<0.01).Conclusions: 1.circ EPSTI1 is up-regulated in gastric cancer tissues,and had a closely correlated with histological grade,TNM stage and lymph node metastasis of gastric cancer patients.2.The introduction of si-circ EPSTI1 into SGC-7901 and MGC-803 cells resulted in increased apoptosis,decreased proliferation,migration and invasion.
