Objective:By observing the expression of Caveolin-1(Caveolin-1,Cav-1)and drug resistance-related proteins(P-gp,GST-?,LRP)in primary liver cancer tissue samples,we can understand the relationship between Cav-1 and related resistance proteins in the body,and then explore the possible mechanism of lipid raft in the formation of multidrug resistance of tumor.We combined with patient follow-up data and clinical pathology information to establish a mathematical model,then used multivariate analysis to determine the prognostic effects of major prognostic indicators for liver cancer prognosis,especially the significance of Cav-1 and drug resistance-related proteins in prognosis,so as to clarify whether high expression of Cav-1 and drug resistance-related proteins is the key factor for poor prognosis of tumor.1.Observed the expression of Cav-1,related resistance proteins(P-gp,GST-?,LRP)and proliferation index(Ki67)in liver cancer surgical specimens to clarify their presence in liver cancer tissue,adjacent liver tissue and far adjacent liver tissue.Then explored their expression patterns2.Investigated the relationship between Cav-1 and related drug resistance proteins in liver cancer tissues to clarify the role of lipid rafts in the fonnation of tumor resistance3.Analyzed the relationship between Cav-1,related resistance proteins(P-gp,GST-?,LRP)and proliferation index Ki67,then combined the follow-up data to explore the relationship between Cav-1 and tumor progression4.Combined with follow-up data,a mathematical model of tumor prognosis was established to evaluate the role of Cav-1,related resistance proteins,cell proliferation activity,and major clinical data such as tumor size,number,grade,and stage in prognosis.Then explored the main risk factors that influence prognosis and their interrelationships.Determined whether high expression of Cav-1 and related resistance proteins is a key factor in determining tumor prognosisMethod:1.120 cases of liver cancer tissue specimens were collected from the pathology department of Zhongshan Hospital affiliated to xiamen University from September 2015 to January 2017.And they were diagnosed as HCC(used the standard "7 points" method).Then,collected the patient's clinical data including the patient's age,gender,tumor size,number,satellite nodules,vascular invasion,tumor grading and staging.2.Immunohistochemistry was used to detect the expression of Cav-1,related resistance proteins(P-gp,GST-?,LRP)and proliferation index Ki67 in liver cancer tissue,adjacent liver tissue and far adjacent liver tissue.They were interpreted using double-blind methods.3.Spearman rank correlation test was used to analyze the correlation between cav-1,related drug-resistant protein and proliferation index Ki67,as well as the correlation between each protein and clinical data.4.A total of 120 patients with liver cancer were followed up to compare the prognosis between patients with high Cav-1 expression and those with low Cav-1 expression.Kaplan-Meier survival analysis,log-rank test and COX risk regression model were used to analyze the effects of each factor on the prognosis of patients.Result:l.The expression of Cav-1,P-gp,GST-?,LRP and Ki67 in hepatocarcinoma tissues was significantly higher than that of adjacent liver cancer tissues in terms of expression intensity and frequency of expression.The difference was statistically significant(P<0.05).There was no significant difference in the expression of adjacent liver tissue and far adjacent liver tissue,the difference was not statistically significant(P>0.05)2.The expressions of Cav-1,P-gp,GST-? and LRP in liver cancer tissues were positively correlated with the expression of Ki67,and the difference was statistically significant(P<0.05).3.The expression of Cav-1,P-gp,GST-? and LRP in liver cancer was positively correlated with the expression of Ki67(P<0.05).4.Vascular invasion and clinical stage of tumor were positively correlated with the expression of cav-1 and related drug resistant protein(P-gp,GST-? and LRP),and the difference was statistically significant(P<0.05).5.Kaplan Meier survival analysis and Log-rank test results showed that vascular invasion,grading,staging and the expression of Cav-1,P-gp,GST-? and LRP in liver cancer tissue affected the prognosis of patients after radical resection of liver cancer risk factors(P<0.05).Patients' age,gender,tumor size,number,satellite nodules and the expression of Ki67 were not significantly correlated with the prognosis of HCC(P>0.05).6.In the COX regression risk model,multivariate analysis showed that the expression of Cav-1 in liver cancer tissues,tumor grading and HCC prognosis were correlated,and the difference was statistically significant(P<0.05).Vascular invasion,clinical staging,P-gp,GST-?,and LRP expression in tumor were not associated with prognosis(P>0.05).In conclusion:1.Cav-1,related resistance proteins(P-gp,GST-?,LRP)and proliferation index Ki67 were highly expressed in liver cancer tissues.2.In liver cancer tissues,the expression of Cav-1 was positively correlated with the expression of related resistance protein.3.Liver cancer tissues with high expression of Cav-1 and drug resistance proteins had high proliferative activity.4.Liver cancer tissues with high expression of Cav-1 and related resistance proteins(P-gp,GST-?,LRP)were more prone to vascular invasion and had higher clinical stage.5.Cav-1 and clinical grading were indicators of poor prognosis of liver cancer.They were independent risk factors for determining the prognosis of liver cancer.
