Purpose:Using genomics to screen differentially expressed genes in gastric cancer and adjacent tissues,analyze the association between the expression levels of related genes and the presence of clinicopathological parameters,and explore new tumor markers for diagnosis and treatment of gastric cancer,providing a new direction for diagnosis and treatment of gastric cancer.Methods:1.This study used the GEO database and TCGA database to analyze the whole gene expression profile of gastric cancer tissues and corresponding adjacent tissues.Some genes were newly discovered,such as: alpha-1-acid glycoprotein(ORM1),type II transmembrane serine protease(HPN),uridine phosphorylase(UPP1),non-metastatic melanoma glycoprotein B(GPNMB).These genes may exhibit differential expression in gastric cancer and adjacent tissues.2.56 patients who were diagnosed with gastric cancer and hospitalized in our hospital from March 2017 to September 2018 were enrolled in the experiment.Real-time quantitative PCR was used to detect the mRNA expression differences of ORM1,HPN,UPP1 and GPNMB in gastric cancer and adjacent tissues.3.Collect clinical data of patients including age,gender,tumor diameter,tumor T stage,differentiation degree,lymphatic metastasis and tumor TNM staging,and conduct statistical analysis to explore the relationship between ORM1,HPN,UPP1,GPNMB expression and clinical data of patients.Results:1.Genomics was used to screen 49 high-expression genes in gastric cancer and 65 low-expression genes in gastric cancer from GEO database and TCGA database.2.Real-time quantitative PCR analysis of the expression of four genes in 56 pairs of gastric cancer and adjacent tissues.Compared with adjacent tissues,GPNMB was highly expressed in 40 cases and were lowly expressed in 16 cases with gastric cancer,the difference was statistically significant(P<0.01),indicating that GPNMB is highly expressed in gastric cancer tissues.Compared with adjacent tissues,ORM1 was lowly expressed in 45 cases with gastric cancer,and 11 cases were highly expressed,the difference was statistically significant(P<0.01),indicating that ORM1 was lowly expressed in gastric cancer tissues.Compared with adjacent tissues,HPN was lowly expressed in 49 cases and high in 7 cases(P<0.001),indicating that HPN was lowly expressed in gastric cancer tissues.UPP1 was highly expressed in 39 cases with gastric cancer,and 17 cases were lowly expressed,the difference was not statistically significant(P>0.05),indicating that UPP1 had no significant difference in gastric cancer tissues.3.According to the results of real-time PCR,combined with the clinical data of patients: GPNMB was highly expressed in gastric cancer tissues(P<0.01),and positively correlated with gastric cancer T stage(P<0.05).The later T stage of gastric cancer,the higher the expression of GPNMB.However,its expression was not significantly associated with age,gender,tumor diameter,degree of differentiation,TNM stage,and lymph node metastasis.ORM1 was down-regulated in gastric cancer tissues(P<0.01).Compared with female patients,ORM1 expression was significantly decreased in male patients(P<0.05).However,There was no significant correlation between ORM1 expression levels with age,tumor diameter,differentiation,TNM stage and lymph node metastasis.HPN was lowly expressed in gastric cancer tissues(P<0.001).There was no significant correlation between HPN expression levels with age,gender,tumor diameter,differentiation,TNM stage and lymph node metastasis.Conclusion:1.Genomics can be used to screen differentially expressed genes in gastric cancer and adjacent tissues.2.The expression of GPNMB in gastric cancer tissues is higher than that in adjacent tissues,and it is related to local invasion of tumors,which may play a certain role in the occurrence and development of gastric cancer.3.The expression of ORM1 and HPN in gastric cancer tissues is lower than that in adjacent tissues,which may play a certain role in the occurrence and development of gastric cancer.4.The UPP1 gene may not be related to the occurrence and development of gastric cancer.
