Acinetobacter haemolyticus which possesses hemolytic activities is identified as an aerobic,nonfermentative,nonmotile gram-negative bacillus.Acinetobacter haemolyticus is responsible for severe nosocomial infections including lung infection,urinary tract infections,bacteremia,endocarditis and meningitis.Through the whole genome sequencing and genomics research of the mechanisms of drug resistance and pathogenesis of A.haemolyticus are with importantly clinical significance.In this study,the whole genome of Acinetobacter haemolyticus TJS01 was sequenced using the third generation sequencing technology,and obtained a circular chromosome and two circular plasmids.Combined with another strain Acinetobacter haemolyticus TJR01,which was sequenced and assembled before,we annotated the genome of two strains from the aspects of gene component,functional genomics,and comparative genomics.The strain TJS01 harbored a circular chromosome of 3,414,636 bp and two plasmids of 6,602 bp and 39,508 bp.Pathogenesis of this strain:the prophage carried a Stx2 related encoding gene,which might be related to bloody diarrhea;thirteen virulence factor related genes,the effects are adhesion,immune escape,stress protein,secretion of the system protein,regulatory protein and oxygen antigen;one siderophore gene cluster,one ectoine gene cluster,one pathogenic island and one metabolic island competitively combine the ferric ion causing the body to reduce immunity and improve the survival rate in high temperature and high salt conditions.The strain TJR01 consisted of a circular chromosome of 3,414,636 bp and two plasmids of 56,602 bp and 39,508 bp;the strain TJR01 expressed two drug resistance genes on chromosome:blaOXA-215 and aac?6'?-Ig.Drug resistance mechanism of this strain:sixteen drug-resistant genes on the plasmid,resistant to antibiotics including acA4,strA,aph?3'?-Via,strB,aph?3'?-Ic,blaPER-1,blaOXA-58,aac?6'?Ib-cr,msr?E?,mph?E?,floR,ARR-3,sul2,sul1,sul1 and tet.The resistance of strain TJR01 is acquired resistance,which might be due to the plasmid-mediated drug-resistant.When contact with antibiotics,gene-mediated metabolic pathway of pathogens changes,then leads to antibiotic inactivation.The genome collinearity of strain TJS01 and TJR01 is well.Comparing the conserved/specific gene,it was found that the two strains had a high degree of matching.Horizontal gene transfer resulted in low localized region of genome.Phylogenetic tree showed that the two strains were close to each other.Acinetobacter haemolyticus TJS01 is the first completed genome in NCBI,and it may be severved as an assembly reference for others.It could provide reliable data for studying the mechanisms of drug resistance and pathogenesis of Acinetobacter haemolyticus.
