Objective: To clarify the expression of Sec10 in the ureter of unilateral congenital ureteropelvic junction obstruction patients,and to explore the correlation between Sec10 and moderate to severe ureteropelvic junction obstruction in children.Materials and methods: 1.Source of experimental specimens: The specimens of the experimental group originated from children with unilateral hydronephrosis caused by congenital ureteropelvic junction obstruction who were hospitalized in our urological ward from January 2015 to December 2018.All the children were examined by color B ultrasonography,intravenous pyelography,CT plain scan,enhancement,three-dimensional reconstruction,and confirmed by operation as unilateral congenital hydronephrosis caused by ureteropelvic junction obstruction.After the approval of the ethics committee of our hospital,the family members of the children were informed and informed consent was signed,the ureter specimens removed during the operation were retained.Thirty-four specimens were retained(n=34).According to SFU classification,the specimens were further divided into moderate group(n=6)and severe group(n=28).The control group consisted of 10 children(n=10)with unilateral nephroblastoma who underwent surgical treatment in our urology department at the same time.All the patients were not treated with chemotherapy before operation,and no invasion of the ureter was found.2.Experimental methods:(1)HE staining: Tissue specimens were fixed,dehydrated,transparent,waxed,embedded,paraffin sections made,HE stained,dehydrated transparent,sealed,and observed under a microscope.(2)Immunohistochemical staining: Immunohistochemical staining,the experimental steps were carried out according to the kit instructions.In immunohistochemical staining,pannoramic viewer software was used to analyze the pictures collected under the microscope.Then quantitative analysis is carried out by using densito quant software in Quant Center.Then each tissue was further analyzed to identify areas(units: pixels)of strong,moderate,weak positive and negative,and positive percentages.At last,H-score was scored.(3)Statistical analysis: SPSS21.0 statistical software was used for statistical analysis.Classified variables are represented by numbers or rates.The continuous variables are expressed by mean±standard deviation.The analysis of variance was used for comparison between groups.Fisher's exact test was used to classify variables.P < 0.05 showed significant difference.Results: 1.Basic clinical data: There was no significant difference in sex,side and BMI among the groups(P > 0.05).2.HE staining results: In the ureteral tissue of the children with obstruction of the group of ureteropelvic junction obstruction,the myofibrillar tissue was obviously increased,the myofibrillar layer was hypertrophic,disordered and sparse,and a large number of coarsened collagen fibers were observed,and the lumen was obviously narrowed.In control group of nephroblastoma,there were no abnormal changes in ureteral tissue and no infiltration of tumor cells.3.Immunohistochemical staining results:Sec10 was positive in moderate group,severe group and control group.Among them,the total positive rate of Sec10 in nephroblastoma group was 98.30%,and H-Score group was 146.90±9.94.The total positive rate of moderate group was 99.62%,and H-Score was 153.92±11.85.The total positive rate of Sec10 in severe group was 98.92%,and that of H-Score was 146.15±9.65.There was no significant difference among the three groups(P > 0.05).4.Relevance analysis: There was no significant correlation between the expression of Sec10 and the severity of ureteropelvic junction obstruction.CONCLUSION: There was no significant difference in Sec10 expression between the ureteral tissue of children with ureteropelvic junction obstruction and that of children with nephroblastoma,and there was no significant correlation between Sec10 expression and the severity of pelvic-ureteral junction obstruction.It is suggested that the expression of Sec10 may not correlate with the occurrence of ureteropelvic junction obstruction.Therefore,this index can not be used as a potential biomarker for the diagnosis of ureteropelvic junction obstruction.
