Objective:To study the expression of Gap junction protein connexin 43 and Ki-67 in the stenotic tissue of Congenital Ureteropelvic Junction Obstruction(UPJO) in Children . in order to elucidate the relationship between both connexin43 and Ki-67 and the pathogenesis of UPJO.Methods:Forty specimens collected from children with UPJO during Anderson- Hynes operation were stained for connexin 43 or Ki-67 by immunohistochemistry respectively.The diagnosis of these patients were confirmed by X-ray imaging and B-mode ultrasonic examination preperatively, 10 biopsies from normal ureteropelvic junction were gathered from postmortem children neither hydronephrosis or neuroal system dieases as control group.Results:Gap junction Cx43 and Ki-67 differently were expressed between the circular muscle and longitudinal muscle in UPJO and control group.The positive rate of Cx43 expression in UPJO group was lower than that in control group(P<0.05) and the expression of Ki-67 in UPJO group was significantly higher than that in control group(P<0.05). There was strongly negative relation between Cx43 and Ki67. The expressions of Ki-67 in UPJO group was closely associated with the degree of hydronephrosis.Conclusion:With the increase of degree of hydronephrosis, the expression of Ki-67 in UPJO showed a rising trend and the expressions of connexin 43 trends to decrease simultaneously.The lack of connexin 43 may lead to the abnormal spontaneous dynamic conduction and cell proliferation in ureteropelvic junction stenosis segment.
