Part1:Association between polymorphism of the G-protein β3subunit C825 T and essential hypertension in Han Chinese population: an updated meta-analysisObjective Meta analysis was performed to determine whether there was a correlation between GNB3C825 T genetic polymorphism and the risk of primary hypertension in Chinese Han population.Methods By searching Pub Med,CNKI,Wan Fang and VIP databases until January 2019,included the related studys by inclusion and exclusion criterion.The correlation was assessed by the odds ratio(OR)and 95% confidence intervals(95%CI)of the five genetic models.A fixed or random effects model was separately adopted depending on the heterogeneity of study.Subgroup or sensitivity analyses were performed to detect the sources of heterogeneity.Publication bias was tested using funnel plots.Results Ten studies were included for meta-analysis,including 3657 patients with hypertension and 3448 individuals in the control group.No significant correlation was found between GNB3C825 T genetic polymorphism and essential hypertension in the meta-analysis of each genetic model: "Tvs C" : OR(95% CI)=1.04[0.97,1.11],P =0.33;"TTvs CC" :OR(95% CI)=1.05[0.90,1.21],P= 0.54;"TCvs CC" : OR(95% CI)=1.08[0.96,1.21],P=0.19;"TT+TCvs CC" : OR(95% CI)=1.13[0.95,1.35],P=0.17;"TTvs TC+CC" : OR(95% CI)=1.01[0.89,1.14],P=0.87.In the further sensitivity analysis,it was found that one study may be the source of heterogeneity of "TT+TCvs CC" genetic model meta-analysis,but excluding the study had no significant effect on the final results.And only one of the 10 included studies was conducted in South Han Chinese population.However,the number of current studies on the Hanpopulation in Southern China is limited,so the influence of genetic variation between the South and North of China on the results cannot be completely excluded.Conclusion GNB3C825 T genetic polymorphism has no significant correlation with primary hypertension in Chinese Han population,but more large sample studies involving Southern Han population are needed to make the conclusion more reliable.Part2:The association of GNB3C825 T genetic polymorphism with the efficacy of amlodipine in the treatment of senile hypertension in Northern AnhuiObjective To investigate the effect of G-proteinβ 3(GNB3)C825T genetic polymorphism on the antihypertensive response to amlodipine in elderly patients with hypertension.Methods Incorporating 100 elderly patients with essential hypertension in Northern Anhui into the study,and give the hypotensor regimens of benzene sulfonic acid amlodipine 5 mg/d for 2 weeks,then record the baseline of blood pressure of patients and the general information about height,weight,age and gender 2 weeks ago.Paying a return visit to measure blood pressure levels after 2 weeks.PCR-direct DNA sequencing was performed to detect the single nucleotide polymorphism(SNP)of GNB3C825 T in patients.The t-test,chi-square test and linear regression analysis were used to assess whether the genetic polymorphism is associated with the antihypertensive effect of amlodipine.Results A total of 83 elderly patients with essential hypertension completed the study,excluding patients who lost to follow up,took drugs without regularity and stopped since side effects.The genetic distribution of the 83 patients satisfies the H-W genetic equilibrium law;Two weeks later the systolic blood pressure(SBP)of patients with CC genotype decreased significantly more than that of T allele carriers(P<0.05),and the diastolic blood pressure(DBP)decreased with the similar rule,but has no statistics significance.And Linear regression analysis was used to adjust the influence of confounding factors such as height,weight,age and gender,and the above conclusions were verified again.Conclusion The genetic polymorphism of GNB3C835 T is related to the efficacy of amlodipine on senile hypertensive patients of Northern Anhui.
