Objective: To investigate whether the variants -930A/G and 242C/T of NAD(P)H oxidase p22phox subunit gene are involved in the pathogenesis of essential hypertension in Chinese Han population from the area of Hubei province.Methods: Genomic DNA was extracted from peripheral blood eukocytes by improved idoic sodium method in both primary hypertension group and normotensive group .the distribution of p22phox gene -930A/G and 242C/T locus polymorphism was identified with polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) in 105 unrelated health contols and 123 patients with primary hypertension. Date of medical history, physical examination were also collected. Automated Biochemical Analyzer was used to measure the plasma lipid level.Results:( 1) For the -930 locus: the allelic frequencies were 0.40 for the A allele and 0.60 for the G allele in hypertension group,0.64 for the A allele and 0.36 for the G allele in normotensive subjects. There was a significant increase in the frequency of G allele gene in hypertention group compared with health one(P<0.01).the frequency of genotypes in the two groups was significant too( P<0.01),and OR=2.67, 95%CI:1.84-3.88(2) For the 242 locus: the frequencies of T allele were 0.16 for hypertension group and 0.07 for control one. the frequencies of CT+TT genotyp in hypertension group and control one were 27.6% and 13.3%, respectively. In the two groups the frequencies of T allele gene and CT+TT genotype were all significant in statistics( P<0.01), and OR=2.48,95%CI:1.31-4.68Conclusions: the results suggest that p22phox gene -930A/G, 242C/T polymorphisms may be involved in the pathogenesis of primary hypertension in Hubei province,and people with G or T allele are easy attacked by hypertension than people with A or C allele; the two polymorphysims maybe do hypertension in coordination.
