Objective:To study the protective effect of different doses ofcycloastragenol on acute renal injury induced by gentamicin in mice and its mechanism.Methods:Thirty healthy male C57BL/6J mice aged 6-8 weeks were randomly divided into blank group,model group,high,medium and low doses of cycloastragenol(800,400,200 mg kg-1)treatment group.The model group and each dose of treatment group were intraperitoneally injected with appropriate amount of gentamicin(100 mg kg-1)for 7 consecutive days.At the same time,the treatment group was given cycloastragenol,the blank group and the model group were given the same amount of physiological saline.On the night after the last administration,fasting and water deprivation were carried out the next day.The orbital blood of mice was collected to detect the contents of creatinine(CRE),urea nitrogen(BUN),reduced glutathione(GSH),superoxide dismutase(SOD)and malondialdehyde(MDA).The left kidney was stained with hematoxylin-eosin(HE)and glycogen(PAS)and observed.The right kidney was homogenized.The contents of GSH,SOD,MDA.MCP-1 and TNF-alpha were measured.Results:Compared with the model group,the treatment group can significantly reduce the contents of CRE,BUN,MDA,MCP-1,TNF-alpha(P<0.05)?and significantly increase the GSH content(P<0.05),SOD activity(P<0.05),but also significantly improve renal tubular necrosis in renal injury.Conclusion:Cycloastragenol has protective effect on gentamicin-induced acute renal injury,and its mechanism may be related to the antioxidant function of cycloastragenol.
