| Objective 1.Study on the inhibitory effect of quercetin on GABA-induced bioelectricity of cortical neurons.2.Study on the effect of quercetin on the food intake of physiological C57 mice.3.Study on the correction of autism-like behavior in BTBR mice with pathological state by quercetin.Methods 1.In vitro cultured cortical neurons,patch clamp technique was used to record the inhibitory effect of different concentrations of quercetin on GABA-induced currents,and the inhibition of different concentrations of GABA-induced currents by the same concentration of quercetin.2.C57 mice were divided into control group,quercetin high dose group(Quercetin50mg/10ml/kg),quercetin medium dose group(Quercetin 25mg/10ml/kg),quercetin low dose group(Quercetin 10mg)/10ml/kg),measuring the food intake of each group of mice.3.C57 mice were divided into control group,quercetin + high-dose muscimol group(Quercetin+muscimol 0.5mg/10ml/kg),quercetin+medium muscimol group(Quercetin+muscimol 0.4mg/10ml/kg),quercetin + low-dose muscimol group(Quercetin+muscimol 0.3 mg/10ml/kg),and the food intake of each group of mice was determined.4.C57 mice were divided into control group and quercetin group.The anxiety level was evaluated in O maze.The brain volume was compared in MRI images.The CTA model was replicated by intraperitoneal injection of Li Cl to evaluate the degree of conditional aversion to each group.5.C57 and BTBR mice were divided into blank administration group(C57-Quercetin),blank control group(C57-Vehicle),administration group(BTBR-Quercetin)and model group(BTBR-Vehicle).Each group was tested in the open filed test,marble burying test,self-grooming,three-chambered social approach test,and the behavioral indicators were recorded.6.BTBR mice were divided into m PFC model group(BTBR-Vehicle-m PFC)and m PFC administration group(BTBR-Quercetin-m PFC),and each group recorded its behavioral indicators in three-chambered social approach test.Results 1.Quercetin can inhibit the GABA-induced current at a concentration of 10umol/L,and the higher the concentration of quercetin,the stronger the inhibition;the 10umol/L quercetin inhibits the GABA-induced current at a concentration of 0.3-300umol/L,and inhibition intensity is independent of GABA concentration.2.Quercetin can reduce the food intake of C57 mice,which is significantly different from the control group(p<0.05),and the dose of 25mg/10ml/kg is the best.3.Quercetin + different doses of muscimol can alleviate the anorexia effect of quercetin on mice.4.There was no significant difference in the behavioral index between the quercetin group in the O-maze test and the CTA test compared with the control group(p>0.05).5.(1)the results of open filed test:There was no significant difference in the movement distance and movement speed between the blank control group and the model group in the open field test(p>0.05),indicating that there was no significant difference in the autonomous exercise ability between C57 mice and BTBR mice;the model group stayed in the central region.The time was shorter than the blank control group(p<0.01),and the residence time of the drug-administered group in the central region was longer than that of the model group(p<0.01),indicating that quercetin can reduce the anxiety level of BTBR mice.(2)the results of marble burying test: the number of buried beads in the model group was significantly higher than that in the blank control group(p<0.05),indicating that the BTBR mice had anxiety-like behavior and repeated stereotypic behavior.The number of buried beads in the drug-administered group was 0,significantly less than Model group(p<0.05),indicating that quercetin can significantly reduce anxiety levels and repeated stereotypic behavior in mice.(3)The results of the grooming: the number of grooming in the model group was more than that in the blank control group,the difference was statistically significant(p<0.05),and the grooming time of the mice in the drug-administered group was less than that in the model group.Academic significance(p<0.001),indicating that quercetin can improve stereotyped behavior and forced-like behavior in BTBR mice.(4)the results of three-chambered social approach test: There was no significant difference between the model group and the time of contact between stranger 1 and empty(p>0.05),and there was no significant difference between the time of contact between stranger 1 and stranger 2(p>0.05),the dose of25mg/10ml/kg.The administration group was exposed to racer 1 for more time than empty(p<0.001),and there was no significant difference in the contact time between stranger 1 and stranger 2(p>0.05).The dose group of 50 mg/10 ml/kg was exposed to stranger 1 There was no significant difference between time and empty(p>0.05),and the contact time of stranger 2was more than that of stranger 1(p<0.05),indicating that 25mg/10ml/kg of quercetin can improve the social interaction ability of BTBR mice,50 mg /10 ml/kg of quercetin can increase the level of social novelty in BTBR mice.6.There was no significant difference between the m PFC model group and the time of exposure 1 and empty(p>0.05).The m PFC administration group was exposed to stranger 1more than empty(p<0.001).The two groups of mice were on strrange 1 and strange 2 There was no significant difference in contact time(p>0.05),indicating that the improvement of autism-like behavior in BTBR mice with quercetin was associated with medial prefrontal lobe.Conclusion 1.Quercetin can non-competitively inhibit GABA-induced bioelectricity,which is positively correlated with quercetin concentration.2.Quercetin can reduce food intake in C57 mice,a process mediated by GABAA receptors,independent of inhibition and conditional taste aversion.3.Quercetin can improve autism-like phenotype in BTBR mice. |
