ObjectiveTo study the effects of Fasudil in Glaucoma.we Established a rabbit model with ocular hypertension by injecting carbomer into anterior chamber.Method:21 rabbits were radomly divided into 7 group,A-original gel group,B-Ag gel group,C-GO gel group,D-Ag+GO gel group,EAcerazolamide+ Ag+GO gel,-Ag+GO +Fasudil gel and G-intraocular hypertension gel.,and the right eyes are experimental group,the left one are control group.We used Goldman tenonometer to test baseline intraocular pressure before injecting.we put medical gel into conjunctiva one days before the intervention,and the three days after the intervention.We measured the IOP twice everyday on 9:00 am and 9:00 pm,after the establishment of carbomer model.If in any group,the IOP were under 22 mmHg,we would inject tthe same amount of carbomer again in every group.we performed Domain Anterior Segment OCT(FDOCT)test after the intervention day one and day six.Then we ethically killed all rabbits at seventh day,tissue specimen taken from eyeballs were made into frozen slice,and we tested the Immunofluorescent ?-SMA.Results:The success rates of IOP model were 88.89%,the peak IOP were appeared at 5-6 day,the peak IOP of Ag+GO +Acerazolamide gel group and Ag+GO +Fasudil gel group were lower than other groups,with statistically significant difference(P < 0.05).other groups IOP level contrast with control group were without statistically significant difference(P>0.05).It suggested that carbomer is an ideal and effective material in building chronic ocular hypertension.fasudil has the effects of reducing ocular pressure.in images of FD-OCT,each group has increased in Corneal thickness(CT)and corneal edema at the sixth day,the Ag+GO + Fasudil group were increased the least,and Ag+GO +Acerazolamide group comes the second,with statistically significant difference(P <0.05),Each groups change in cornea were similar with the clinical manifestation of Glaucoma.A postive results of Immunofluorescent ?-SMA expression were showed in controlgroup,while negative results were observed in the contrast group,it is indicated that the fibrillation of conjunctiva ocurred,and wound scar healing happened..Ag+GO +Fasudil group showed a lower positive rate than other groups,and with statistically significant difference(P<0.05).The result suggested that Fasudil has an Anti-fibrosis effect,also Sub-conjunctival delivery of medicine is an effective and safe wayConclusions:Carbomer is an ideal and effective method in building an animal model with chronic ocular hypertension.Fasudil has the effects of reducing IOP and Anti-fibrosis.Sub-conjunctival delivery of medicine is a new method in opthalmology.
