Objective:Tissue microarray and immunohistochemistry method was used to detect the expression level of autophagy-related genes ATG3 and Ambra1 in gastric cancer tissues,cancer adjacent tissues and normol gastric tissues,and analyze the correlation between its expression and clinical pathological factors.Methods:It collected 40 cases of gastric adenocarcinoma surgically resected from the Second Hospital Affiliated of Nanhua university,and collected 40 cases of cancer adjacent tissue and 20 cases of normal gastric tissue.By using tissue microarray analysis,each sample was made by tissue microarray.Using immunohistochemistrymethod to detect the expression level of ATG3 and Ambra1 in each sample.The correlations of ATG3 and Ambra1 expression with clinical pathological factors such as sex,age,TNM staging,differentiation degree were analyzed.Results:1.The expression difference of ATG3 and Ambra1 in gastric cancer tissues,cancer adjacent tissues and normol gastric tissueswas statistically significant.2.The positive expression rates of ATG3 were 77.50%,70.00% and 40.00% in gastric cancer tissues,cancer adjacent tissues and normal gastric tissues,and the difference among the three groups were statistically significant(p<0.05).The comparison between the three groups showed that the difference was not statistically significant between cancer group and adjacent group(p>0.05),and the differences were statistically significant between cancer group and normal group(p<0.005),adjacent group and normal group(p<0.05).The positive expression rates of ATG3 in lymph node metastasis group and non-lymph node metastasis group were 95.00% and 60.00%,and the difference between the two groups was statistically significant(p<0.01).The positive expression rate of ATG3 in gastric cancer was not correlated with age,gender,tumor invasion depth,TNM stage and differentiation degree(p>0.05).3.The positive expression rates of Ambra1 were 50.00%,32.50% and 15.00% in gastric cancer tissues,cancer adjacent tissues and normal gastric tissues,and the difference among the three groups were statistically significant(p<0.05).The comparison between the three groups showed that the difference was statistically significant between cancer group and normal group(p<0.01),and the differences were not statistically significant between cancer group and adjacent group(p>0.05),adjacent group and normal group(p>0.05).The positive expression rates of Ambra1 in lymph node metastasis group and non-lymph node metastasis group was 70.00% and 30.00%,and the difference between the two groups was statistically significant(p<0.05).The positive expression rates of Ambra1 were 29.41% and 65.22% in T1-T2 group and T3-T4 group,and the difference between the two groups was statistically significant(p<0.05).The positive expression rate of Ambra1 in gastric cancer was not correlated with patient's age,sex,TNM stage and differentiation degree(p>0.05).4.In 31 cases of ATG3 positive gastric adenocarcinoma samples,there are positive expression of Ambra1 in 19 cases,and in 9 cases of ATG3 negative gastric adenocarcinoma samples,Ambra1 expression was negative in 8 cases.There was a significant correlation between ATG3 and Ambra1 expression in gastric cancer(p<0.01).Conclusion:1.ATG3 and Ambra1 are highly expressed in gastric cancer,and it may promote lymph node metastasis of gastric cancer through autophagy.2.The expression of Ambra1 in gastric cancer was correlated with tumor invasion depth.
