Background: lung cancer morbidity and mortality are among the highest in the world,but as the curative effect of existing treatments is limited,new treatment methods are urgently needed to be found.Human-derived xenografts(PDX)are increasingly being used in models for the development of new drugs for lung cancer.This kind of direct transplantation of the patient's tumor tissue into the immunodeficient mouse model has been proved by many studies to retain the original tumor heterogeneity,molecular pathological features,and closer to the clinical,and become a drug sensitivity test,a preclinical study of new drugs.And an important platform for the discussion of drug resistance mechanisms.ethaselen(1,2-[bis(1,2-benzisoselenazolone-3(2H)-ketone)]et hane,BBSKE)has been shown to inhibit the inhibition of Thioredoxin Reductase(TrxR)activity A variety of tumor growth,and has been demonstrated in a variety of in vitro cell tests BBSKE inhibition of lung cancer,breast cancer,colorectal cancer,melanoma and other malignant tumor cell proliferation.In the cell-line-derived xenograft(CDX)model,BBSKE combined with Cisplatin(DDP)has been shown to inhibit lung cancer growth.This study was the first to use PDX to verify the effect of BBSKE on lung cancer growth.Objective: This study is based on the PDX model to investigate the effects of BBSKE/selenium ethaselen(BBSKE-SE)on the growth of squamous cell lung carcinoma.Methods: The surgical specimens of lung squamous cell carcinoma of Hunan Cancer Hospital were collected and transplanted into the immunodeficient nude mice to establish a PDX model of lung cancer.The success of the modeling was verified by HE staining.The experimental mice were further divided into 6 groups: 1.DMSO 2.DMSO + DDP(2.5mg/kg)3.BBSKE(72 mg/kg)4.BBSKE + DDP 5.BBSKE-SE(72 mg/kg)6.BBSKE-SE + DDP.The tumor volume change curve was drawn according to the growth of the tumor,and the curve of the body weight change of the mouse was drawn.Mainly to explore the value-added effect of BBSKE and BBSKE-SE on squamous cell lung carcinoma.Results:(1)Hematoxylin-eosin staining(HE)staining method confirmed that the tumor type of PDX model was consistent with the original tumor,suggesting that PDX modeling was successful.(2)In the PDX model,BBSKE can inhibit the proliferation ofsquamous cell lung carcinoma.(3)BBSKE-SE can inhibit the proliferation ofsquamous cell lung carcinoma,and the effect is more significant than BBSKE.(4)Therewas no statistical difference in body weight changes between mice in all groups,suggesting that mice can tolerate drugs.Conclusion: 1.PDX modeling success;2.PDX model to verify that BBSKE can inhibit squamous cell lung carcinoma appreciation;3.BBSKE-SE can inhibit squamous cell lung carcinoma appreciation,and the effect is more significant than BBSKE.
