Objective:The genes highly expressed in epithelial ovarian cancer tissues(CENPF,ZWINT)were screened by bioinformatics technology,and the protein expression levels of CENPF and ZWINT genes in epithelial ovarian cancer tissues and normal ovarian tissues were verified by immunohistochemistry.Explored the correlation of the protein expression levels of CENPF and ZWINT with the development of ovarian cancer,various clinical indicators and prognosis.Explored potential of CENPF and ZWINT genes as biomarkers for the diagnosis and prognosis in epithelial ovarian cancer.Methods:1.By analyzing the mRNA microarray datasets GSE14407,GSE6008 and GSE36668 from the Gene Expression Omnibus database,the genes CENPF and ZWINT with significant expression differences associated with ovarian cancer were screened out.2.Collected paraffin sections from 73 patients with primary epithelial ovarian cancer admitted to the Department of Gynecologic Oncology,Hunan Cancer Hospital from January 2011 to April 2013 and 25 normal ovarian tissues.The expression level of ZWINT and CENPF genes were examined by immunohistochemistry.3.Analyzed the correlation of ZWINT and CENPF gene expression levels with prognosis and clinicopathological features and explored its clinical significance in the screening and diagnosis of epithelial ovarian cancer.Results:1.CENPF was lowly expressed in normal ovarian tissue(25 cases).CENPF was highly expressed in 31 of the 73 epithelial ovarian cancer tissue.The positive expression rate was 42.5%.The expression of CENPF in epithelial ovarian cancer tissue was significantly higher than that in normal ovarian tissue(P<0.01).The expression of ZWINT was low in both normal ovarian tissue and epithelial ovarian cancer tissue,and there was no difference in expression.2.The expression of CENPF in the moderately and poorly differentiated epithelial ovarian cancer group was significantly higher than that in the highly differentiated group.The expression of CENPF was significantly different from the grade of differentiation in epithelial ovarian cancer tissue(P <0.05).3.The expression of CENPF in the group with ascites ? 500 ml was significantly higher than that with ascites <500ml.The difference between expression of CENPF and ascites was statistically significant(P<0.05).4.There was no significant difference in CENPF expression level from age,tumor grade,omentum and lymph node metastasis in patients with epithelial ovarian cancer.(P> 0.05).5.Survival analysis on 73 patients with ovarian cancer was performed by Kaplan-Meier.The overall survival(OS)of patients with high CENPF expression was significantly lower than that of patients with low CENPF expression.The 5-year and 7-year overall survival rates of patients with high CENPF expression were 38.5% and 31.5%,the median survival time was 54 and 54 months.The 5-year and 7-year overall survival rates of patients with low CENPF expression were 67.4% and 48.4%.The 7-year median survival time was 74 months.The difference was statistically significant(P<0.05).Multivariate analysis was performed by COX proportional hazard regression model which showed that the expression of CENPF is a prognostic factor of epithelial ovarian cancer(P<0.05,HR=1.964,95%CI:1.006-3.834).Conclusion:1.The expression of CENPF was high in epithelial ovarian cancer tissues;2.The expression level of CENPF was negatively correlated with the degree of differentiation of epithelial ovarian cancer and positively correlated with the amount of ascites.3.CENPF expression is an important prognostic factor for epithelial ovarian cancer.The overall survival rate of patients with high CENPF expression was lower than that with low CENPF expression.
