With breast cancer being a complicated disease with the high heterogeneity,there are lots of classification methods on breast cancer.However,most of these methods are not exactly accurate and have little guidance at clinical level,especially for triple negative breast cancer which has high malignancy and poor prognosis precise with no effective treatment available so far.Finding subtyping gene markers feasible for clinical cancers treatment has been a crucial task for improving the identification,diagnosis and treatment of breast cancers.Here we explore the role of FOXA1,SOD2 and IL6 in characterizing triple negative breast cancers and its driving mechanisms.We used bioinformatics combined with molecular experimental validation.Through in vitro examination of the expression pattern at both transcriptional and translational levels,analysis of gene expression data from public databases using breast cancer cells and clinical samples,we systematically compared the role of FOXA1,SOD2 and IL6 in identifying triple negative and non-triple negative type of breast cancers.We analyzed and validated the interaction among FOXA1,SOD2 and IL6 by predicting transcriptional binding using public databases,chromatin immunoprecipitation assay and luciferase reporter assay.We studied function of FOXA1,SOD2 and IL6 by doing lots of cell biology experiments.The results showed that FOXA1 under-expression can lead to increased malignancy and transcriptionally suppress the expression of SOD2 and IL6.Together with SOD2 and IL6,FOXA1 can influences apoptosis,migration and proportion of stem cells,suggesting very important roles of FOXA1,SOD2 and IL6 on breast cancer progression.We are the first to systematically address the significance of FOXA1,SOD2 and IL6 in triple negative breast cancer identification as biomarker genes both in cell and in tumor level,and elucidate the mechanism at the molecular level.Our discoveries of the current biomarker genes improve the precision on characterizing breast cancer heterogeneity,which is conducive to early clinical diagnosis and prognostic judgement of patients.Function researches on FOXA1,SOD2 and IL6 make it possible to turn them into clinical therapeutic targets on triple negative breast cancer.
