Objective This study contains two parts.The fitst part aims to investigate the Piezol expression level in para-carcinoma tissues compared with osteosarcoma tissues.In the second part,we try to explore the Piezol expression level in cells treated with curcumin,and to analyse the effect of curcumin combined with Piezol siRNA on cell migration and apoptisis.Methods Clinical samples of osteosarcoma from patients who were pathologically diagnosed as osteosarcoma and para-carcinoma were collected for the immunohistochemical staining to evaluate the expression level of Piezol.In addition,U2-OS cells were cultured and treated with curcumin and Piezol siRNA.QRT-PCR,western blot and RNA-sequence gene profiling were allpied to analyse the expression of Piezol.Ca2+ measurements were applied to detact the functional changes of Piezol ion channels.Also,scratch-wound healing assay and Annexin V/PI stainning were observed to measure the influences of the treatment on migration and apoptosis.The data were analyzed using SPSS 17.0.Results In our study,as showed from the IHC staining,Piezol is significantly upregulated in OS specimens compared to the para-carcinoma tissue.In vitro,we noticed that the piezol expression and protein were significantly downregulated in cells treated with curcumin,and the mRNA sequencing indicated that Piezol was downregulated in cells treated with curcumin comparing with the control group.Next,we specifically downregualted Piezol and found that the group of curcumin collaborating with Piezol silencing presented the strongest inhibnition on cell migration and Piezol channel' s function.Also the highest apoptosis rate was noticed in this group.Conclusion All results above suggest that Piezol exhibit a close connection with tumor development and the anticancer effect of curcumin may rest on the regulation of Piezol.Specifically downregualted Piezol could enhance the cells sensitivity to curcumin.Further work investigating will be necessary to obtain a better understanding of the functions of Piezol in the development of osteosarcoma.
