Objective: To investigate the expression of the miR-124in osteosarcoma MG-63treated with curcumin and their effects on cell growth as well as associatedmechanisms.Methods: We evaluated the expression levels of miR-124in osteosarcoma of twogroups (Curcumin treated group and DMSO control group) by qRT-PCR. The cellkilling effects of miR-124in osteosarcoma were measured by MTT and Woundhealing methods. We used the Western blot and luciferase reporter assay to identifydirected target gene of miR-124.Results: Curcumin decreased the osteosarcoma cells MG-63growth in andose-dependent manner by MTT. Transfection of miR-124mimics for12h,24h,48hrespectively, MTT assay showed that the cells growth were inhibited significantlycompared with the control group (P <0.05)(The killing effect rates were47.9%,58.5%or67.1%, respectively), and it was time-dependent; Transfection of miR-124mimics for24h, cells wound healing assay showed that miR-124could significantlyslowed down the scratch wound healing of the MG-63, the groups were significantdifferent (P <0.05); Luciferase reporter assay confirmed that SPHK1(sphingosinekinase, Sphingosine kinase1, SPHK1) was a direct target gene of miR-124; Westernblot showed that the expressions of SPHK1protein were inhibited by miR-124.Conclusions:1. Curcumin inhibit the osteosarcoma cells MG-63growth by up-regulating theexpression of miR-124.2. SPHK1is the target gene of miR-124in osteosarcoma cell line MG-63.3. miR-124can inhibit osteosarcoma cell growth by down-regulating the expressionof SPHK1.
