| Objective:To investigate the effect of Bushen Tiaogan Recipe on the expression of SOX9 and Runx2 in rats with knee osteoarthritis(KOA),and to determine the mechanism of articular cartilage protection?In order to improve the clinical efficacy of traditional Chinese medicine,provide more experimental applications for prevention and treatment of KOA.Method:1 Establishment of KOA animal model:A KOA rat model was established using modified Hulth modeling.2 Experimental group:Rats were divided into sham operation group,KOA model group(blank group),KOA model?Bushen Tiaoganfang group(Chinese medicine group),KOA model + glucosamine group(Western medicine group),12 in each group.The rats were sacrificed at different time points to take cartilage tissue for correlation detection.3 Gavage treatment3.1 According to the human and rat dose conversion formula,prepare 1.9g of crude medicine/ml of Bushen Tiaogan Decoction,13.9mg/ml concentration of glucosamine sulfate(Viekel)solution.3.2 From the 13th week after modeling,the rats in the Chinese medicine group and the western medicine group were given Bushen Tiaogan Decoction and Weiguli solution for intragastric administration.4 HE(Hematoxylin-eosin staining,HE)The knee articular cartilage tissues of the right hind limbs of each group were stained with HE at different time points.The OARSI score was used to evaluate the therapeutic effect of Bushen Tiaogan Recipe on cartilage repair.5 WB(Western Blot,WB)The cartilage tissues of the right hind limbs of each group were taken at diff'erent time points.The expressions of Runx2,SOX9 and ?-catenin were detected by WB method,and the effects of Bushen Tiaogan Recipe on its expression were analyzed.Result:1.The OARSI scores of the KOA rats in the Chinese medicine group and the western medicine group were significantly decreased after the intragastric administration of Bushen Tiaogan Recipe and Glucosamine Sulfate,but the difference was statistically significant(P<0.05).2.The expression of SOX9 protein in the articular cartilage tissue of KOA rats decreased,and Runx2 increased.However,the expression of normal cartilage tissue did not change significantly.3.Bushen Tiaogan Recipe can up-regulate the expression of SOX9 in the cartilage tissue of KOA rats,and down-regulate the expression of Runx2.After 4 weeks of treatment with Bushen Tiaogan Recipe,the expression of SOX9 protein is increased and the expression of Runx2 protein is decreased,Statistical significance(P<0.05).4.?-catenin was negatively correlated with the expression of SOX9 and positively correlated with the expression of Runx2.Conclusion:The effects of Bushen Tiaogan Recipe on the expression of OARSI,SOX9 and Runx2 in articular cartilage of KOA rats were observed and compared.The following conclusions were obtained:(1)Bushen Tiaogan Recipe is superior to glucosamine sulfate capsule in the protection of articular cartilage in KOA rats.(2)Bushen Tiaogan Recipe plays a therapeutic role by up-regulating the expression level of SOX9 protein,promoting chondrocyte proliferation and self-repair,inhibiting the degeneration and absorption of chondrocytes and cartilage matrix;(3)SOX9,Runx2 dynamic t.emporal and spatial balance is to maintain joints a key factor in the normal morphology of cartilage. |
