Research Background and Objective:Esophageal cancer?EC?is a common malignant tumor of the digestive tract. In China, the incidence of EC accounted for the third and the mortality became the fourth which was mainly esophageal squamous cell carcinoma?ESCC?.The research investigates the expression of cluster of differentiation 151?CD151?and extracellular regulated protein kinase1/2?ERK1/2?and the presence of vasculogenic mimicry?VM?in ESCC.The correlations which among with the clinicopathological features of ESCC,the presence of VM,and the relationship between the presence of VM and prognosis were analyzed further.Materials and Methods:General Materials:100 cases of paraffin-embedded ESCC tissues samples from January 2012 to June 2012 in the First Affiliated Hospital of Bengbu Medical College were enrolled in the observation group and 50 cases of normal esophageal tissues as the control group. Methods:The expression of CD151 and ERK1/2 in ESCC was detected by ElivisionTM plus method. The presence of VM in esophageal cancer was detected by CD34/PAS double staining.Statistical Analysis:All data were statistically analyzed by using SPSS 16.0statistical software. In the ESCC tissues, the expression of CD151,ERK1/2,and VM in the observation group and the control group, and the correlations between with the clinicopathological features were analyzed by?2 and Spearman correlation analysis. If P<0.05,there is a significant difference between them, and the difference is statistically significant.Survival analysis was performed by using the Kaplan-Meier method.Results:1.The positive rates of CD151,ERK1/2 and the present rate of VM in the observation group were significantly higher than those in the control group??2=70.936P=0.000,?2=21.520 P=0.000,?2=8.566 P=0.003?.In the observation group, the expression of CD151 protein correlated with the depth of invasion, lymph node metastasis, clinical stage, degree of differentiation??2=19.143 P=0.000,?2=11.968 P=0.001,?2=4.996 P=0.025,?2=14.855 P=0.001?.The expression of ERK1/2 protein was correlated with lymph node metastasis, clinical stage and differentiation of ESCC??2=10.470 P=0.001,?2=12.007P=0.001,?2=12.181 P=0.002?.In the observation group, the expression of ERK1/2 in CD151 positive group was significantly higher than that in ESCC's CD151 negative group, and there was a positive correlation between them?r=0.451,P=0.000?.In the observation group, there was a positive correlation between CD151 and ERK1/2 and VM expression?r=0.299 P=0.002,r=0.318 P=0.001?.2.Kaplan-Meier survival curve showed that the 5-year survival rates of VM group and non-VM group were 10.3%and 50.7%,respectively.The median overall survival?OS?of VM group and VM-free group was 16 months and 51.5 months, respectively. The overall 5-year survival rate of this group was 39.0%,and the median OS was 41months?P<0.05?.COX multivariate regression analysis demonstrated that the depth of invasion, clinical stage, and VM may be independent factors in the 5-year survival rate of patients with esophageal squamous cell carcinoma?P<0.05?.Conclusion:CD151 and ERK1/2 are crucial in the occurrence and development of esophageal squamous cell carcinoma, and may affect the development, metastasis and prognosis of esophageal squamous cell carcinoma by promoting the formation of VM.
