Objective:This study collected D1,D3,D5,and D7 cerebrospinal fluid samples from patients with a SHA(primary subarachnoid hemorrhage),extracted m RNA and sequenced the genome,screened for differential genes,and analyzed differentially expressed genes through bioinformatics.The technology understands the biological function of differentially expressed genes and speculates on the polarization state of Ar cells after SAH.Methods:By collecting D1,D3,D5,and D7 cerebrospinal fluid samples from patients with a SAH,m RNA was extracted and sequenced by sequencing,sequencing of gene data quality statistics,reference genome alignment analysis,transcript assembly and new transcript prediction,known genes.Differential expression,differential gene function analysis,screening differential genes and analyzing the biological functions enriched by differentially expressed genes,detecting the differential gene expression levels at different time after SAH,and inferring the polarization state of Ar cells.Results:By sequencing the m RNA genome,we found that the number of reads of the two samples D1 and D3 in the same patient was 987 differential genes,of which 752 were up-regulated,77%,and 235 were down-regulated,accounting for 23%;D3,D5 two samples of the number of reads,after screening to obtain 1210 differential genes,of which 859 up-regulated genes,accounting for 71%,down-regulated 351 genes,accounting for 29%;D5,D7 two samples of the number of reads,after screening A total of 2016 differentially expressed genes were found,of which 1226 were up-regulated,accounting for 61%,and 790 were down-regulated,accounting for 39%.By further screening the above three pairs of differentially expressed genes,we selected differentially expressed genes related to the polarization state of Ar cells after a SAH,and predicted the functional information of differentially expressed genes by biological protein technology,and found that IL-12(interleukin)-12),IL-18,,IL-27,TGF-? and other cytokines related to M1 polarization increased with the development of disease time,while IL-10 showed low expression levels.Conclusions:1.Ar cells as intrinsic mononuclear macrophages in the subarachnoid space,Ar cells will function differently in polarization state after a SAH,and participate in the late inflammatory reaction.2.Ar cells tend to be in the MI polarization state after aSAH.
