Objective:Study the expression of PD-1,PD-L1 and FOXP3 in pancreatic ductal adenocarcinoma(pancreatic ductal adenocarcinoma,PDAC)tiss ue and paracanc-erous pancreas,combined the expression of PD-1,PD-L1 and FOXP3 in pancreatic cancer with the clinicopathological parameters,to find out the clinical significance and relation of both three proteins.Medthods:45 cases of PDAC tissues and corresponding normal pancreatic tissues adjacent to the carcinoma,which Formalin-fixed and paraffI ne-embedded were collected from the department of pathology of the First Affiliated Hospital of Nanchang University from January 2012 to September 2017.using Immunohistochemistry to detect the expression of PD-1,PD-L1 and FOXP3 in PDAC and corresponding adjacent pancreat ic tissues,SPSS 19.0 software was performed to statistical analysis of experimental data.Results:1.PD-1 was mainly expressed in cytostroma,and the positive expression rates in PDAC and corresponding adjacent pancreatic tissues were respectively 71.11%(32/45)and 37.78%(17/45).The positive expression rate of PD-1 in PDAC group was significantly higher than that in adjacent pancreatic tissue(P< 0.05).The expression of PD-1 correlated with histological grade,nerve infiltration,lymph node metast asis and TNM stage in PDAC(P < 0.05),but not related to age,sex,tumor locat ion and tumor size(P > 0.05).2.PD-L1 was mainly expressed in cytoplasmic,and the positive expression rates in PDAC and corresponding adjacent pancreatic tissues were respectively 64.44%(29/45)and 42.22%(19/45).The positive expression rate of PD-L1 in PDAC group was significantly higher than that in adjacent pancreatic tissue(P < 0.05).The expression of PD-L1 was related to histological grade,nerve infiltration,lymph node metastasis and TNM stage(P < 0.05),but not related to age,sex,tumor location,tumor size(P > 0.05).3.FOXP3 was mainly expressed in cytonucleus,and the positive expression rates in PDAC and corresponding adjacent pancreatic tissues were respectively 80%(36/45)and 13.33%(6/45).The positive expression rate of FOXP3 in PDAC group was significantly higher than that in adjacent pancreatic tissue(P < 0.05).The expression of FOXP3 was related to PDAC nerve infiltration and lymph node metastasis(P < 0.05),but not related to age,sex,tumor location,tumor size,histological grade and TNM stage(P > 0.05).4.Spearman correlation analysis showed that in PDAC,the expr ession of PD-1 was positively correlated with PD-L1 expression(r = 0.542,P < 0.05),PD-1 and FOXP3 expression were not correlated(r= 0.130,P > 0.05),and there was a positive correlation between the expre-ssion of PD-L1 and FOXP3(r=0.398,P < 0.05).Conclusion:Both PD-1/PD-L1 signaling pathway and FOXP3 play a negative role in immune regulation,which is closely related to the occurrence and development of pancreatic cancer,and can promote the invasion and metastasis of pancreatic cancer.In addition,the abnormal expression of the signal pathway and FOXP3 can make pancreatic cancer have more malignant biological behavior.There is no direct correlation between FOXP3 and independent PD-1,it is related to PD-L1.Detection both PD-1,PD-L1 and FOXP3 may contribute to the evaluation of the prognosis of pancreatic cancer and provide a theoretical basis for adjuvant treatment of pancreatic cancer.
