AIM To investigate the relationship between the expression of WT1, IGF-IR and apoptosis in human pancreatic duct adenocarcinoma.Methods Immunohistochemical analysis was used to detect the expression of WT1, IGF-IR in 49 cases of human pancreatic duct adenocarcinoma and 15 cases of normal pancreatic tissue. Apoptosis of pancreatic duct adenocarcinoma was estimated by terminal deoxynucleotidyl transferase mediated dUTP nick end labeling(TUNEL)method and apoptosis index (AI) was calculated.Results In normal pancreatic tissue the positive rates of WT1, IGF-IR were 26.67%(4/15), 40.00%(6/15), and in pancreatic duct adenocarcinoma the positive rates of WT1, IGF-IR were 71.43%(35/49), 77.55%(38/49)which were significantly higher than those in normal pancreatic tissue (P<0.05). There was a positive relationship between the expression of WT1 and IGR-IR in pancreatic duct adenocarcinoma(r=0.385, P<0.05). The apoptosis index in the normal tissue and pancreatic duct adenocarcinoma were 0.63±0.21 and 5.93±4.18, there was a significant statistical difference between the normal group and pancreatic duct adenocarcinoma group (P<0.05). The AI increased with the rising of tumor differentiation degree in pancreatic duct adenocarcinoma. The AI of the IGF-IR positive groups was lower than those of the negative groups in pancreatic duct adenocarcinoma.Conclusion The apoptosis was inhibited by the overexpression of IGF-IR in pancreatic duct adenocarcinoma and the overexpression of WT1, IGF-IR and apoptosis may play an important role in the genesis and progression of pancreatic duct adenocarcinoma.
