Objective:By detecting the expression level of interleukin35(IL-35) in the mucosal tissues of chronic rhinosinusitis with masal polyps(CRSwNP),and studying the cytokines and preoperative CT of TH1/TH2/TH17 The correlation between the scoring system,the visual analog scale score,and IL-35 was used to investigate the possible role of IL-35 in the pathogenesis of CRSwNP.Methods:1.The expression of two subunits of IL-35(IL-12P35 and EBI3) in the tissues of CRSwNP group(30 cases) and control group(17 cases) was detected by immunohistochemistry(HC).2.The expression level of IL-35 in tissue homogenate was measured by enzyme-linked immunosorbent assay(ELISA).3.Real-time quantitative polymerase chain reaction(q-PCR) was used to detect the expression levels of IL-12P35,EBI3,IFN-?,IL-5,IL-17 A and FOXP3 mRNA in tissues.The correlation between them.4.The sinus CT(Lund-Mackay) and visual analogue scale(VAS) were collected from the above 30 patients with nasal polyps,and their correlation with IL-12P35 and EBI3 was analyzed.Results:1.IHC results showed that the expression of IL-12P35 and EBI3 in CRSwNP was lower than that in the normal group,and it was important that there was a significant correlation between the two.2.The level of IL-35 protein in CRSwNP tissue homogenate was lower than that in the normal group,and the difference was statistically significant.The results of q-PCR showed that the expression levels of IL-12P35,EBI3 and FOXP3 mRNA in CRSwNP group were significantly lower than those in the control group(P<0.05).The relative expression of various inflammatory factors IFN-?,IL-5 and IL-17 A mRNA.The level was higher than that of the control group(P<0.05).The expression levels of IL-12P35 and EBI3 mRNA in polyp patients were significantly correlated with various cytokines.4.The expression levels of IL-12P35 and EBI3 were negatively correlated with sinus CT(Lund-Mackay)and visual analogue scale(VAS).Conclusion:The expression of IL-35 in nasal polyps is closely related to various cytokines and is closely related to the severity of the disease,indicating that IL-35 may play a key role in the pathogenesis of CRSwNP.IL-35 may become a new target for the treatment of CRSwNP.
