| 1 clinical study1.1 purpose To study the distribution characteristics of TCM syndromes in patients with severe and very severe COPD.1.2 method Literature inquiry,combined with the tutor’s diagnosis experience,referring to "TCM diagnostics" for syndrome differentiation and treatment,four diagnosis and reference,and related knowledge of "TCM internal medicine",designed questionnaire survey and formulated TCM syndrome questionnaire for patients with severe and extremely severe COPD.1.3 the results Among the 120 patients with severe and extremely severe COPD,the first three syndrome types were lung kidney qi deficiency syndrome(31.7%),phlegm turbidity obstruction to lung syndrome(23.4%),phlegm heat stagnation to lung syndrome(20.8%).1.4 the conclusion Severe and extremely severe syndromes of COPD are mostly manifested as deficiency of lung and kidney qi with blood stasis,and deficiency and excess.2 experimental research2.1 purpose By studying the effects of shenqi huatan prescription containing serum on the expressions of Rho Am RNA,ROCKI/IIm RNA and mypt-1 m RNA in ASMCs,the correlation between COPD airway remodeling and the prevention and treatment of Rhoa/ROCK signaling pathway was discussed,providing sufficient scientific and theoretical basis for clinical treatment of COPD.2.2 method TThe serum of shenqi huatan prescription and blank control serum were prepared.Then,airway smooth muscle cells(ASMCs)were isolated and cultured.Finally,ASMCs with good growth were selected and treated with DMEM overnight to achieve uniform cell growth to G0 phase.The cells were randomly divided into six groups: blank group,model group(20% blank serum),blocker Fasudi(3mmol/L),blocker Fasudi(3mmol/L)+SQHTF group(5%,10%,20%).Three parallel holes shall be set for each group.After 24 hours,tnf-a(10ng/m L)was added to all groups except blank,and the indicators to be tested were further cultured for 24 hours.The expression levels of ASMCs Rho Am RNA,ROCKI/IIm RNA and mypt-1 mrna in each group were detected by rt-qpcr.2.3 the results 2.3.1 Compared with blank control group,the rest of the group Rho Am RNA airway smooth muscle cells,Rock Ⅰ m RNA,Rock Ⅱ m RNA,MYPT 1 m RNA expression were significantly higher than that of normal group,difference has statistics meaning(P < 0.001);2.3.2 Compared with model group,the treatment group rats Rho Am RNA airway smooth muscle cells,Rock Ⅰ m RNA,Rock Ⅱ m RNA,MYPT 1 m RNA expression are lower,including Rock Ⅰ m RNA has no statistical significance(P > 0.05),more than between groups(P < 0.001),statistically significant;2.3.3 Compared with Fasudil group,SQHTF + Fasudil Rho Am RNA airway smooth muscle cells in each group,Rock Ⅰ m RNA,Rock Ⅱ m RNA,MYPT 1 m RNA expression reduced,the difference was statistically significant(P < 0.05 or P < 0.01 or P < 0.001);2.3.4 Comparison between various SQHTF + Fasudil group,10% and 20% SQHTF + Fasudil group rats Rho Am RNA airway smooth muscle cells,Rock Ⅰ m RNA,Rock Ⅱ m RNA,reducing the most significant MYPT 1 m RNA expression,difference have statistical meaning(P<0.001);Compared with 10% SQHTF + Fasudil group,20% SQHTF + Fasudil group rats Rho Am RNA airway smooth muscle cells,Rock Ⅰ m RNA,Rock Ⅱ m RNA,MYPT 1 m RNA expression is reduced,but not significant,is not statistically significant(P>0.05).2.4 the conclusion SQHTF drug serum reduced the expressions of Rho Am RNA,ROCKI/IIm RNA and mypt-1 m RNA in ASMCs,inhibited airway inflammation and participated in airway remodeling,providing a new idea for the future treatment of COPD. |
