Study On Tiainzhusan Constructed With Increasing Herbs According To Tiarnma And New Formula Design

Objective:Data mining method was used to mine the herbal pairs of formulas containing Gastrodia elata;and the expert consultation method was used to investigate the commonly used combination of herbal medicine containing Gastrodia elata,whichcould be used to prevent and treat stroke.It is a feasible method to use the network pharmacology to explore the relationship between "herbal medicine-ingredients-targets-pathways" for the study of compatibility mechanisms and formulas design.Methods:With the help of R language arules package,the association rules of Chinese herbal formulas containing Gastrodia elata were analyzed,and the high frequency herbal pairs were screened out according to the Support and Confidence.The high frequency herbal pairs were subjectively evaluated by Delphi expert consultation method,and the common clinical herbal pairs were supplemented.Then the core herbal pairs containing Gastrodia elata were obtained with the subjective and objective weight analysis method.The R language arules package was used to analyze the association rules of formulas including.Gastrodia elata The high frequency herbal pairs were selected according to the ranking of Support and Confidence;Delphi expert consultation method was used to subjectively evaluate the high-frequency herbal pairs and and the commonly used clinical herbal pairs were supplemented.Subjective and objective weight analysis methods were used to obtain the core herbal pairs containing Gastrodia elata.The herbal pairs containing Gastrodia elata were analyzed by network pharmacology,including screening of small molecular active substances,predicting herbal candidate targets,analyzing biological functions of candidate targets,analyzing interaction between disease targets,analyzing interaction between candidate targets and disease targets,and drawing network of "traditional Chinese medicine-targets-pathways".Finally,the New Formula Design of TianZhuSan was carried out by screening herbs with better medicinal properties,and the molecular mechanism of TianZhuSan was analyzed by network pharmacology.Results:1.Association rules analysis obtained 27 groups of high-frequency herbal pairs containing Gastrodia elata,which were "Gastrodia elata-Radix sileris","Gastrodia elata-Scorpion","Gastrodia elata-Musk","Gastrodia elata-Rhizoma typhonii","Gastrodia elata-Bombyx batryticatus","Gastrodia elata-Rhizoma seu Radix Notopterygii","Gastrodia elata-Cinnabar","Gastrodia elata-Arisaema","Gastrodia elata-Ginseng","Gastrodia elata-Monkshood","Gastrodia elata-Chinese ephedra","Gastrodia elata-Cinnamon","Gastrodia elata-Radix Angelicae Pubescentis","Gastrodia elata-Pinellia ternata","Gastrodia elata-Calculus bovis","Gastrodia elata-Radix achyranthis bidentatae","Gastrodia elata-Asarum","Gastrodia elata-Radix Aucklandiae","Gastrodia elata-Realgar",etc.2.Based on Delphi expert consultation method,12 groups of commonly used herbal pairs containing Gastrodia elata were supplemented,including “Gastrodia elata-Rhizoma acori graminei”,"Gastrodia elata-Pueraria lobata","Gastrodia elata-Cassia seed","Gastrodia elata-Fructus viticis","Gastrodia elata-Concha haliotidis","Gastrodia elata-Chrysanthemum","Gastrodia elata-Acanthopanax Senticosus","Gastrodia elata-Notoginseng","Gastrodia elata-Fructus Amomi","Gastrodia elata-Dried tangerine peel","Gastrodia elata-Sappanwood","Gastrodia elata-Concha Ostreae”.3.The objective and subjective weight analysis method was used to screen out the herbal pairs containing Gastrodia elata with higher weight value,including “Gastrodia elata-scorpion”,“Gastrodia elata-Bombyx batryticatus "," Gastrodia elata-Chrysanthemum "," Gastrodia elata-Ligusticum wallichii "," Gastrodia elata-Pueraria lobata "," Gastrodia elata-Concha haliotidis "," Gastrodia elata-Rhizoma acori graminei "," Gastrodia elata-Rhynchophylla "," Gastrodia elata-Fructus viticis "," Gastrodia elata-Achyranthes bidentata”,etc.4.Network pharmacology was used to analyze seven groups of herbal pairs:“Gastrodia elata-Chrysanthemum","Gastrodia elata-Ligusticum wallichii","Gastrodia elata-Pueraria lobata","Gastrodia elata-Rhizoma acori graminei","Gastrodia elata-Ramuli et spina uncariae","Gastrodia elata-Trillium tschonoskii” and “Gastrodia elata-Borneol”.The results showed that there were more active small molecule compounds except for Trillium tschonoskii,which showed good intestinal absorption and brain permeability.There were 17 overlapping targets between components of “Gastrodia elata","Chrysanthemum","Ligusticum wallichii","Pueraria lobata","Rhizoma acori graminei","Ramuli et spina uncariae","Trillium tschonoskiiBorneol "and stroke targets,which correspond to 107 chemical components and 15 major metabolic pathways.5.The candidate targets of Tianzhusan(TZS)plus borneol or Tianzhu powder plus Gouteng or Tianzhu powder plus Chuanxiong have good interaction with stroke targets.There were more identical targets in three groups of formulae and stroke targets,such as ACE,ACE2,APP,FOLH1,HIF1 A,MMP9,etc.These targets have the functions of regulating blood pressure,promoting learning or improving memory,etc.Conclusion:Data mining association rules is an objective and effective method to mine the compatibility law of formulas.Based on the improved Delphi expert consultation method,we investigated the clinical application of herbal pairs containing Gastrodia for stroke prevention and treatment,and grasped the commonly used clinical herbal pairs containing Gastrodia in the prevention and treatment of stroke from the subjective level,remedied for the deficiency of data mining mechanical calculation.The prevention and treatment mechanism of Gastrodia elata on apoplexy was explained from microcosmic point of view by using network pharmacology technology,which embodied the complex network interaction characteristics of multi-component,multi-target and multi-pathway between herbal medicine and herbal pairs.It further provides research methods for the design of new Chinese Medicine formulas.
