With the rapid advancement of modern science and technology,radioactive radiation is emerging in all walks of life,and people will always be exposed to radiation in their daily lives.However,some of the existing drugs with better anti-radiation therapeutic effects are difficult to promote and apply due to their toxic and side effects.Therefore,it is necessary to find a radioprotectant with good anti-radiation effect and low toxic side effects from plants and their natural components.It must be done.In this paper,Kunming mice were used as a model to explore the anti-radiation function and mechanism of mulberry leaf powder and its active ingredients.An X-ray mice injury model(4.0 Gy)was established to observe the effect of mulberry leaves powder on radiation-induced mice.The organ coefficient,spleen lymphocyte proliferative capacity,bone marrow DNA content,bone marrow micronucleus rate serum and liver antioxidant enzyme activities,and malondialdehyde content were measured.The results showed that compared with the radiation control group,the organ coefficient,spleen lymphocyte proliferation,bone marrow DNA content and antioxidant enzyme activity of the mulberry leaf powder group were significantly increased,and the micronucleus green and malondialdehyde content decreased significantly.The results showed that mulberry leaf powder can effectively protect mice from radiation damage.Different doses of radiation(4.0,6.0,8.0,10.0 Gy)were used to establish the survival model of radiation-injured mice.The organ coefficient,spleen lymphocyte proliferation ability,tissue section,blood routine and bone marrow DNA content of each group were detected.,bone marrow micronucleus rate serum and liver antioxidant enzyme activity and malondialdehyde content and other indicators to compare the anti-radiation function of mulberry active ingredients.The results showed that the anti-radiation effect of Morin was the best in survival rate,organ coefficient,spleen lymphocyte proliferation ability,blood routine,bone marrow micronucleus rate,bone marrow DNA content,antioxidant enzyme activity and malondialdehyde content.The last DNJ is mulberry leaves powder.The anti-radiation molecular mechanism of Morin,DNJ and mulberry leaves powder was explored by RT-PCR.The results showed that the up-regulation of apoptosis-relatedgene FAS(inhibition of apoptosis)and the down-regulation of ATK and Caspase2 genes together inhibited apoptosis and reduced radiation damage.Both hematopoietic and cytokine-related genes improve the radiation resistance of the body by improving the hematopoietic system.The combination of CD23 up-regulation and IL-1 up-regulation results in a certain recovery of hematopoietic function in mice after irradiation.The activated expression of the antioxidant genes GCLM,NQO1,and HO-1 significantly increases the body's antioxidant capacity,thereby eliminating a large amount of free radicals generated by radiation to protect the body from radiation.
