The method of MTT assay was used to evaluate the minimal effective dose of Dendrobine and diallyl sulfide mcf-7,respectively.FCM was used to assess the synergy effect between Dendrobine and diallyl sulfide.Apoptosis was studied by Annexin V-FITC/PI staining analysis and flow cytometer.Changes of mitochondrial transmembrane potential were observed by the means of JC-1 fluorescence confocal.Via Western blotting we probed the expression of caspase-3 protein,Bcl-2 protein and Bax protein.Result:With MTT assay,it was found that the inhibition of allyl sulfide on mcf-7 was more effective than that of dendrobine,and the minimal effective concentrations of the two chemicals and their combination were 001mg/ml,0.2 mg/ml,and 0.lmg/ml+0.2mg/ml,respectively.The early detection of apoptotic rate was recorded in dendrobine treatment group,which was 10.58%?11.03%?16.69%.The early apoptosis rate was recorded in diallyl sulfide treatment group,which was 33.96%?40.73%?51.46%.The comprehensive effect of the two drugs was significant,even the apoptotic rate was as high as 73.84%.MCF-7 cells cycle was arrested in G2/M phase by diallyl sulfide,Dendrobine arrested the cell cycle in G1/M phase.In the MCF-7 cells treated with combination of the two drugs,the most obvious polarization phenomenon was found based on the clear changes in mitochondrial membrane potential.The results demonstrated that apoptosis inducing factors was released into the cytoplasm upon the two drugs were added,then the cell death mechanism was started,and which subsequently triggered a cascade of reactions.WB detection of Bax,Bcl-2,CDK1,CyclinB1,caspase-3 protein,showed that diallyl sulfide regulated CDK1 and CyclinB1 protein kinase leading to inhibiting the proliferation of tumor cells in G2/M phase.Whereas dendrobine down regulated the expression of Bcl-2 protein,while up regulating the expression of Bax,which activated expression of the execution of apoptosis factor caspase-3,finally resulted in apoptosis of MCF-7 cells.In conclusion,the data identified that there was a significant interaction effect between the two drugs,and it implied that the combination of the two drugs could achieve promising results as an effective therapy to tackle the breast cancer,multi-target integration effect leads to its terminal pharmacological action based on correspondence of prescription with the syndrome of traditional Chinese medicine.
