Objective:By comparing with western medicine and routine acupuncture,we observed the effect of "tiaoshenchangqing sanliujiu" acupuncture on the behavior, the contents of CCK and VIP in serum and brain tissues of constipation rats with PD,and the relevant mechanism of action was discussed. Methods:Parkinson's model of SD rats was prepared by injection of6-hydroxydopamine single striatum with two targets. After successful PD modeling, loperamide was used to model PD with constipation. The rats were randomly divided into the normal group, sham operation group, model group, western medicine group, routine acupuncture group and "tiaoshen" acupuncture group. After 14 days of treatment, behavioral evaluation was conducted.Tissue samples were selected at corresponding time points.Results:1.Compared with the normal group, the changes of behavioral indicators in the model group showed significant differences(P<0.05),and the contents of CCK and VIP were significantly reduced in the model group, showing extremely significant differences(P<0.01).2.Compared with the model group, the behavioral indicators of the western medicine group were significantly different(P< 0.01),and the increase of CCK and VIP in the brain tissue and serum of the rats was significantly different(P<0.05).There were significant differences in the behavioral indicators of the rats in the conventional acupuncture group(P<0.05),and there were significant differences in the increase of CCK and VIP in the brain tissue and serum of the rats(P<0.05).The behavioral indexes of the “ tiaoshen” acupuncture group rats showing extremely significant differences(P<0.01),and CCK and VIP in the brain tissue and serum of the rats showing extremely significant differences(P<0.01).3.Compared with the conventional acupuncture group, the behavioral indexes of the western medicine group showed extremely significant differences(P<0.01),and the contents of CCK and VIP in the serum and brain tissues of the rats showed extremely significant differences(P<0.01).Compared with the western medicine group, the behavioral indicators of the“ tiaoshen” acupuncture group were not statistically significant(P>0.05),and the contents of CCK and VIP in the serum and brain tissues of the rats were significantly different(P<0.05).4.Compared with the conventional acupuncture group, the behavioral indexes of the“ tiaoshen” acupuncture group were significantly different(P<0.05),and the contents of CCK and VIP in the serum and brain tissues of the rats showing extremely significant differences(P<0.01).Conclusion:1.In improving the behavior of PD rats with constipation,"tiaoshenchangqingsanliujiu" acupuncture method can achieve the effect of Western medicine treatment, and is superior to conventional acupuncture treatment.2.The three treatments have effects on increasing the contents of CCK and VIP,and the "tiaoshenchangqingsanliujiu" acupuncture method has the best effect, followed by western medicine treatment, and then conventional acupuncture treatment.3.One of the mechanisms of "tiaoshenchangqingsanliujiu" acupuncture may be through regulating the contents of brain-gut peptide CCK and VIP.
