ObjectiveTo study the effect of IGF-1 on invasion and metastasis of lung cancer A549 cells and its possible signaling mechanism.MethodsA549 lung cancer cells were cultured and divided into control group,IGF-1 group,IGF-1 + LY294002 group,and inhibitor LY294002 group.After 48 hours of culture,cell proliferation was detected by MTT proliferation test.The migration ability of cells was tested by cell scratch test.Transwell test was used to detect cell invasion.Western blot was used to detect the expression of AKT,p-AKT,MMP-9,MDM2 and E-cad proteins.ResultsCompared with the control group,IGF-1 significantly promoted the proliferation migration and migration(p<0.05).Western blot showed that IGF-1 promoted the high expression of MMP-9,MDM2 and p-AKT protein and decreased the expression of E-cad protein(p<0.05).AKT inhibitor LY294002 can inhibit the above-mentioned effect of IGF-1.ConclusionIGF-1 may up-regulate MMP-9,MDM2 protein and reduce E-cad protein expression through AKT pathway to affect the invasion and metastasis of lung cancer A549 cells.
