[Background] Gastric cancer is one of the common malignant tumors of digestive system.Metastasis and invasion are the main causes of death.In China,most patients with gastric cancer have metastasized at the time of diagnosis.Because gastric cancer mainly metastases through lymph nodes,Therefore,timely and accurate judgement of lymph node metastasis is very important for the choice of treatment and prognosis evaluation of gastric cancer.At present,there is still lack of effective measures for early metastasis of gastric cancer detection.In recent years,aptamers have become a research hotspot in the field of cancer diagnosis and treatment.So far,there is no report on DNA aptamers targeting highly metastatic gastric cancer cells.In this study,cell-SELEX technique was used to screen aptamers for highly metastatic gastric cancer cells.[Objective] To Screen aptamers for highly metastatic gastric cancer cells HGC-27 by cell-based systematic evolution of ligands by exponential enrichment(cell-SELEX).[Methods] Use highly metastatic gastric cancer line HGC-27 as target cells and non-metastatic line AGS as negative cells to obtain aptamers that specifically bind to HGC-27 cells by using cell-SELEX.Flow cytometry and Laser confocal microscopy was used to analyze the biological propertiesof selected aptamers.[Results] 1.We sucessfuly obtained one target aptamer named LW-25 which can bind highly metastatic gastric cancer cell line with high affinity and specificity.2.The binding site of LW-25 to target cells may be cell surface protein.3.The aptamer can bind to highly metastaic gastric cancer tissues with high sentisively and specificity.[Conlusions] In this study,cell-SELEX technique was used to successfully screen aptamer LW-25 in highly metastatic gastric cancer cells.Preliminary identify its binding site may be cell membrance protein.It lays a foundation for early metastasis diagnosis,precise localization and targeted therapy of metastatic gastric cancer..
