Background: Bladder cancer is a common malignant tumor of the genitourinary system.With the improvement of detection methods,the incidence rate in China is gradually increasing in recent years.Due to the characteristics of local invasion and metastasis of bladder cancer,the patients need to follow the risk according to the degree of risk.The follow-up usually includes regular cystoscopy,urine cytology and urinary tract imaging examination?The intensity and invasiveness of these tests may lead to complications that endanger the patient's quality of life and create a financial burden.In addition to active clinical treatment,screening diagnosis,prognosis assessment,and recurrence monitoring of bladder cancer have always been an important auxiliary link for treatment.Molecular mechanisms for the occurrence and development of bladder cancer,and new methods for monitoring prognosis have been the focus of urology research.TPD52L1(D53)protein is a member of tumor protein family D52(tumor protein D52,TPD52),and is considered to be a new therapeutic target for breast cancer,closely related to cell proliferation and cell cycle control.TPD52L1 was found to be highly expressed in various tumors,but its expression level and physiological role in urinary tumors have not been reported.Objective: The purpose of this study was to detect the expression of TPD52L1 in bladder urothelial carcinoma by immunohistochemistry(two-step method),and the influence of TPD52L1 on the prognosis of patients with bladder urothelial carcinoma was studied by analyzing the clinicopathological data and follow-up results.Methods: Retrospective analysis was performed on the clinical data of bladder cancerpatients admitted to the department of urology of the first affiliated hospital of anhui medical university from 2013 to 2017.According to the established screening criteria,72 cases of bladder urothelial carcinoma were selected as the study subjects.After the pathological section of the patient was taken,the expression of TPD52L1 was detected by immunohistochemistry(two-step method).The correlation between the expression level of TPD52L1 and the prognosis of bladder urothelial carcinoma was analyzed using SPSS22.0.Kaplan-meier method was used to draw the survival curve,calculate the average survival time and survival rate.Univariate analysis using Log-rank test,multivariate analysis using Cox proportional hazard model.Results: Immunohistochemical results showed that 94.4% of bladder urothelial carcinomas had TPD52L1 expression,while peripheral normal urothelial cells were generally negative.Using chi-square test to analyze the data of each group,it was found that the expression level of TPD52L1 was correlated with whether the tumor broke through the whole layer of the bladder(P=0.001).Univariate survival analysis by Kaplan-Meier showed that the disease specific survival of bladder urothelial carcinoma was significantly correlated with the expression intensity of TPD52L1(P<0.001,Log Rank test).multivariate survival analysis using Cox regression showed that the high expression of TPD52L1 was an independent risk factor for disease specific death in bladder urothelial carcinoma(HR3.093,P=0.033).Conclusion: No expression of TPD52L1 was found in normal bladder urothelium,but expression of TPD52L1 was found in most bladder urothelial carcinomas.Moreover,with the increase of its expression level,its disease-specific mortality increased,high expression of TPD52L1 is an independent risk factor for the prognosis of bladder urothelial carcinoma.
