Purpose: To evaluate the feasibility of low-frequency pulsed electromagnetic field to promote the recovery of neurological function after delayed repair of peripheral nerve injury,and to explore the expression of BDNF and VEGF protein in the delayed repair model of peripheral nerve injury in rats.Methods: Sixty rats were used to construct a peripheral nerve injury model.The model rats were randomly divided into low-frequency pulsed electromagnetic field treatment group(n=30)and nerve injury delayed repair group(n=30)by random number method.Surgical model(sham operation group).The model rats in the low-frequency pulsed electromagnetic field treatment group were stimulated by low-frequency pulsed electromagnetic field after sciatic nerve disconnection and delayed repair.The nerve injury delayed repair group only performed sciatic nerve disconnection and delayed repair;the sham operation group only exposed the sciatic nerve without disconnection.Three groups of rats underwent functional studies at 4 and 8 weeks postoperatively,and the sciatic nerve function index was measured.After the rats were sacrificed,the sciatic nerve tissue was taken,and the VEGF and BDNF protein contents were determined by immunohistochemistry after sectioning,and semi-quantitative comparison was performed.At the 8th week after surgery,the sciatic nerve tissue protein was extracted and WB was performed to detect the levels of VEGF and BDNF protein.Semi-quantitative comparison was performed and statistical analysis was performed.Results:(1)Rat sciatic nerve function index results showed that the sciatic nerve index correction factor of the low frequency pulsed electromagnetic field treatment group was significantly higher than that of the nerve injury delayed repair group at 4 and 8 weeks after delayed repair.There was a statistically significant difference in the function index of rat sciatic nerve between different groups(P<0.05).(2)Western Blot results showed that the expression of brain-derived neurotrophic factor and vascular endothelial growth factor in the sciatic nerve of the low-frequency pulsed electromagnetic field treatment group was significantly higher than that of the nerve injury delayed repair group at 8 weeks after delayed repair.The difference of rat sciatic nerve function index was statistically significant(P<0.05).(3)The immunohistochemical results of sciatic nerve tissue showed that the rats in the low-frequency pulsed electromagnetic field treatment group were treated at 4 and 8 weeks after delayed repair.The number of sciatic nerve cell regeneration and brain-derived neurotrophic factor expression were significantly higher than those of the nerve injury delayed repair group.There were significant differences in the sciatic nerve function index between the different groups(P<0.05).Conclusions: The nerve injury delayed repair model was established by a method of nerve suture after a week of sciatic nerve was cut off.The measurement of sciatic nerve function index,immunohistochemistry and WB experiment showed that the low frequency pulsed electromagnetic field can promote the nerve function recovery after delayed repair of the peripheral nerve injury model.
