Background Spinal cord injury is a common CNS injury,and the pathophysiological mechanism is very complex.Spinal cord injury can be divided into primary injury and secondary injury,in which axonal demyelination is one of the important changes of secondary injury.OPCs are progenitor cells of myelin regenerated OLs.Mi RNAs has been shown to play a critical role in the differentiation of OPCs.As a noninvasive and safe biophysical therapy,PEMF has been shown to improve symptoms in neurodegenerative chronic demyelinating diseases,but it is unclear whether PEMF can affect the differentiation of OPCs and its mechanism.Objective The PEMF was constructed to explore its role and mechanism in the differentiation of OPCs.Methods PEMF were constructed and OPCs were randomly divided into PEMF group and NPEMF group.In order to clarify the effect of PEMF on the differentiation of OPCs,the relative expression of specific markers in different differentiation stages of OPCs was detected by immunofluorescence and western blot after 3 days,7 days,14 days and 21 days of exposure,respectively.RT-q PCR was used to further detect the expression of mi RNAs related to OPCs differentiation after exposure.Then,RT-q PCR was used to detect the relative expression of genes associated with candidate mi RNA,which may play a negative regulatory role in the differentiation of OPCs.Finally,after transfection of mi RNA,the relationship between mi RNA and target genes and the effects of PEMF and mi RNA on the differentiation of OPCs were explored under the exposure of PEMF and NPEMF.Results Compared with NPEMF group,the relative expression of OPCs specific marker NG2 in PEMF group was lower in early(3 days,7 days)and late(14 days,21 days)stages.The relative expression of Galc,a specific marker of immature OLs,was higher than that of NPEMF group in the early stage,but lower than that of NPEMF group in the late stage.The relative expression of MBP,a specific marker of myelin regeneration OLs in early and late stage,was higher than that in NPEMF group.Compared with the two groups,PEMF significantly increased the relative expression of mi R-219-5p and decreased the relative expression of Lingo1.Under PEMF,mi R-219-5p targeted Lingo1 and cooperated with PEMF to promote the differentiation of OPCs.Conclusion PEMF promotes the differentiation of OPCs through upregulating mi R-219-5p in vitro,which provides a potential therapeutic strategy for the treatment of Spinal cord injury.
