Target: To study the expression levels of miR-375 in metabolic tissues,organs and serum of diet-induced obese mice at different life stages.And associated that with the gut microbiota of the corresponding stage mice.To explore the early diagnostic value of miR-375 in obesity induced by gut microbiota imbalance.Method: Forty-eight C57BL/6 mice were randomly divided into two groups:HF(high-fat diet)and LF(low-fat diet),and raised in standard animal animals under standard conditions.Free to take water and eat.Body weight and feces were collected at the same time every week.Six mice in each group were sacrificed every 3 weeks,and the serum,the upper small intestine,the middle small intestine,the lower small intestine,the muscle,the liver,the pancreas and the fat were collected at the same time.The expression level of miR-375 was measured by real-time PCR.The composition of the gut microbiota of mice was detected by 16 SrDNA amplicon sequencing.Then analyze the relationship between them.RESULTS: 1)With the increase of feeding time,the difference between the weight of the HF and the LF became larger.After 12 weeks of feeding,the weight of the HF was 37.56±3.16 g,and the weight of the LF was 29.09±2.98 g.The HF weighed 29%more than the LF.The obesity model was successfully modeled because the mice in the HF weighed more than 20% of the LF.2)The expression levels of miR-375 were changed in the samples of the two groups at different stages of feeding,and the expression levels of miR-375 were different in different tissues(P<0.05).There was a difference in the expression of miR-375 between the two groups(P<0.05).The expression levels of miR-375 in the pancreas of the HF were higher than those in the LF(P<0.05).Conversely,the expression of miR-375 in the serum of the LF was higher.Different expression of miR-375 in different small intestine segments.3)There was no significant difference in gut microbiota.diversity between the two groups(P>0.05),but the distribution and number differences were statistically significant(P<0.05).The gut microbiota of the two groups of mice was dynamically changed with the feeding time;The Firmicutes is the most abundant in the gut microbiota of mice.Conclusions: 1)60% of high-fat diets can establish a c57bl/6 obesity model in a relatively short period of time.2)The gut microbiota is different in C57BL/6 mice at different life stages.HF can change gut microbiota,and there is an important relationship between changes of gut microbiota and obesity.3)MiR-375 is expressed differently in different tissues.Specific expression of miR-375 in serum and pancreas and miR-375 is of great value in the early diagnosis of obesity and metabolic syndrome.MiR-375 has different expression levels in different tissues of the body.And it is specifically expressed in serum and pancreas.MiR-375 plays an important role in the early diagnosis of obesity and metabolic syndrome.
