| Objective To investigate the expression of KLK11 in ovarian cancer andthe correlation between its expression and prognosis; to study thesignificance ofKLK11 in tumorigenesis ,metastasis and epidermis alternationfrom RNA levels and protein levels; to supply an experimental basis for thegene diagnosis and the evaluation of prognosis of ovarian cancer.Methods All these cases were divided into three groups: malignant ovariancancer group, benign ovarian tumor group, normal ovarian tissue group. Theexpression of hK11 protein in three groups was examined by means ofimmunohistochemistry and the expression level of the KLK11 gene mRNA inthese cases was tested by reverse transcription-polymersase chain reaction(RT-PCR). The correlation between its expression and tumour clinical stages,histopathology grouping was also studied.Results (1) The expression of KLK11 gene mRNA and hK11 proteinincreased gradually in three groups; (2)the expression of KLK11 gene mRNAwas significantly lower in ovarian cancer than that in benign ovarian tumorand normal ovarian tissue(P=0.022,P=0.021); the expression of hK11 wassignificantly lower in ovarian cancer than that in benign ovarian tumor andnormal ovarian tissue(P=0.016,P=0.013); (3) the expression of KLK11 genemRNA and hK11 in serous type was higher than that in other types of ovariancancer(P=0.045,P=0.012); (4)the expression of KLK11 gene mRNA andhK11 in early stage of ovarian cancer (â… /â…¡stage) was significantly higher than that in late stage (â…¢/â…£stage) (P=0.002,P=0.010); (5) there was nocorrelation between the expression level of KLK11/hk11 and pathologicalstages of ovarian cancer(P=0.510,P=0.747). There was significant correlationbetween the expression level of KLK11/hk11 and clinical stages, histologicaltypes of ovarian cancer.Conclusions The expression of KLK11 gene mRNA and hK11 was existedin ovarian cancer, benign ovarian tumor and normal ovarian tissue. Theexpression of KLK11 mRNA and hK11 was correlated with theclinicopathological features of ovarian cancer and can be a potentialmonitoring biomarker for the diagnosis and prognosis. |
