Background and objectiveNowadays,breast cancer is one of the most common malignant tumors among women all over the world.It has seriously threatened the physical and mental health of women,and the age of patients tends to be younger.In China,the incidence of breast cancer rose sharply.Tumor recurrence and distant metastasis after treatment are the main causes of death.However,the molecular mechanisms involved in tumorigenesis,progression,recurrence and metastasis are still unclear.Therefore,to search of its potential prognostic molecular markers and therapeutic targets,deep study of the molecular mechanism of the occurrence and development of breast cancer is necessary,which is helpful to improve the level of diagnosis and treatment and it is helpful to extend the survival time and improve the life quality of patients.Cancer occurrence has miscellaneous causes,includes the activation of proto-oncogenes and the inactivation of tumor suppressor genes.Activation of a proto-oncogene can occur through a point mutation,gene amplification,or gene translocation.These mutations can alter the DNA copy number and gene function at various locations of a specific genome.It was reported that copy number alterations(CNAs)affect gene expression,which may be a critical component of tumor progression.Detecting CNAs may provide a basis for clinicians and scientists and find out new biomarkers or signaling pathways for therapeutics development or early interference to prevent cancer.With the rapid development of biological and computer technology,mass data has been emerging.More and more public databases have been established and applied for studying the genomic alterations in human cancers.A large number of studies in the literature suggest that PCMT1 gene may be involved in the occurrence and development of tumor.PCMT1 gene may have a potential role in the mechanism of breast tumor,which is expected to be the biomarkers of future targeted therapy.In this study,the expression and clinical significance of PCMT1 gene in breast cancer were preliminarily verified through in-depth mining and analysis on Oncomine.?LinkedOmics and Kaplan-Meier Plotter databases,providing ideas for the follow-up basic research of our group.MethodsThrough extensive reading of relevant literatures,we speculated that PCMT1 gene may be related to relevant signaling pathways and may be involved in the occurrence and development of malignant tumors.After selecting the research target,we logged in the Oncomine database and input the screening conditions in the database interface.In all these data sets,we considered that the experimental results were statistically significant when P value<0.05.Therefore,we selected 12 data sets and extracted the mRNA expression level of PCMT1 gene on the Oncomine database.In addition,a meta-analysis was performed on these data sets on the Oncomine database website.Then we logged in the Linkedomics database and register first to gain access to the database.Screen conditions were selected step by step in the database retrieval interface,and the data related to PCMT1 gene expression in breast cancer and survival prognosis were downloaded.Kaplan-meier method was used to calculate the relationship between PCMT1 expression and prognosis of breast cancer patients.At last,we logged into Kaplan-meier Plotter database to download the survival curve of breast cancer patients.When P value<0.05,significant statistical difference was considered.ResultsTo confirm the role of PCMT1 gene in breast cancer,we first systemically analyzed mRNA expression between normal tissues and various tumor tissues on the Oncomine database.Then,we could find a total of 197 studies on the expression of PCMT1 gene in different types of tumors.According to the screening conditions in this study,twelve data sets about the mRNA expression of PCMT1 gene in breast cancer tissues and normal tissues were selected,including 1018 samples in total.In these twelve study data sets,PCMT1 expression in breast cancer tissues was higher than that in normal tissues(P<0.05).A secondary analysis of the data set of on these twelve studies found that the expression level of PCMT1 in breast cancer tissues was higher than that in normal tissues compared with the control group(P<0.05).In addition,we analyzed the data from LinkedOmics and Kaplan-Meier Plotter database and found that the overall mortality of breast cancer patients with high PCMT1 mRNA expression was higher,while the prognosis of breast cancer patients with low PCMT1 mRNA expression was better(P<0.05).ConclusionPCMT1 gene encodes protein methyltransferase,which is involved in biologlical processes including protein repair,synaptic transmission,cell matrix interaction and apoptosis.Besides,PCMT1 gene may be related to the occurrence and development of cancer.PCMT1 expression is associated with poor prognosis in a variety of malignancies.PCMT1 gene is highly expressed in breast cancer tissues and is associated with the prognosis of breast cancer patients.We may speculated that PCMT1 may play a role as an oncogene in the occurrence and development of breast cancer and is expected to become a new target and a new marker for the prognosis of breast cancer.
