Objective:To explore the expression of CSRP1(cysteine and glycine rich protein1)in colorectal cancer tissues,explore related gene databases,explore the relationship between CSRP1 and colorectal cancer metastasis and prognosis,and provide a new marker basis for clinical diagnosis and treatment of colorectal cancer.Method:The experimental process mainly involves the following three aspects:1.Mass spectrometry techniques to determine differentially expressed protein profiles.Methods: 1)Select 20 colorectal cancer patients(pathological type adenocarcinoma)and take their cancer tissues and corresponding normal tissues,extract the proteins in the above tissues,and prepare them into a mixture of polypeptides.2)Screening differential expression of proteins in the above two tissues by chromatography and mass spectrometry.3)Analysis of the above proteins to determine differentially expressed protein profiles.2.Review the relevant literature,select CSRP1 as the target protein of this study,and verify the expression of this protein in cancer tissues and normal tissues of colorectal cancer patients by Western blot.3.Using bioinformatics,explore and analyze related databases,and use software to organize information about CSRP1 in the database to study its relationship with colorectal cancer.Result:1.Through mass spectrometry and analysis,the results showed that there were 23 differentially expressed proteins in colorectal cancer tissues and their corresponding peritumoral tissues,including 17 up-regulated proteins and 6 down-regulated proteins.The target protein of this experiment was one of the expressed proteins is down-regulated.2.Western blot results showed that the expression of CSRP1 in colorectal cancer tissues was significantly lower than that in normal tissues around the cancer.3.Analysis and integration of Oncomine database showed that the expression of CSRP1 gene in colorectal cancer tissues was lower than that in normal cancer tissues;exploring the Cbioportal database,the mutation rate of CSRP1 gene was low with 1.11%;use COSMIC database analysis the mutation type of this gene in colorectal cancer,we found that the proportion of missense mutations was the highest,reaching 73.08%;the proportion of nonsense mutations was the lowest,accounting for 9.62%;the synonymous mutation was at middle,accounting for 21.15%;Cbioportal database analysis showed CSRP1 co-expressed genes in colorectal cancer were ranked according to relevance.The top ten were TAGLN,CNN1,MRCPRF,DACT3,KCNMB1,HSPB7,ACTG2,LMOD1,and MYLK.The Centoscape software was used to analyze CSRP1 protein byCluego and Cluepedia.Among the related pathways,the most important is Dilated Cardio Myopathy(DCM),accounting for 24.56%,followed by Muscle Cell Development,accounting for 22.81%.Conclusion:1.CSRP1 is down-regulated in colorectal cancer tissues,and its expression in cancer tissues is lower than that in normal tissues.2.The gene encoding CSRP1 has a low mutation rate in colorectal cancer,which is suitable for subsequent gene therapy research,and this gene is closely related to multiple signal transduction pathways of colorectal cancer,which is worthy of further exploration and clinical diagnosis of colorectal cancer.This research also provides new evidence and ideas for treatment.
