According to the theory of traditional Chinese medicine,the syndrome of qi-blood deficiency is a holistic and comprehensive disease.It is caused by chronic illness,overwork,excessive blood loss or senile,which leads to the decline of the function of qi-blood or the function of organs.The syndrome of qi deficiency is more common in the body's immune function,causing damage to immune organs,immune cells,immune molecules and other functional levels;the study of blood deficiency syndrome mainly focuses on the impact of the body's hematopoiesis and immune system,from peripheral blood cell counts,bone marrow hematopoietic system and hematopoietic microenvironment,thymus and spleen cell apoptosis.AnShenBuNao oral liquid has the functions of replenishing marrow and qi and nourishing blood,strengthening brain and soothe the nerves,and is often used for kidney deficiency,fatigue,forgetfulness,insomnia,nerves,weakness and other diseases.Objective:According to the function of An Shen Bu Nao oral liquid,the model of qi-blood deficiency in mice was established.The anti qi-blood deficiency effects of An Shen Bu Nao A and B were compared with the old and new techniques,anti-fatigue,and sedative and hypnotic.Methods: 60 male Kunming mice weighting 20-22 g were randomly divided into 6 groups in each group,namely blank control group(distilled water),model control group(distilled water),An Shen Bu Nao A 5,10 m L/kg dose group and the An Shen Bu Nao B 5,10 m L/kg dose group were administered intragastrically once a day for 10 consecutive days.On the 4th,6th,and 8th day after administration,the model control group and the drug-administered group bleed 0.25 m L/10 g in the tail vein.On the 5th,7th,and 9th day,the model control group and the drug-administered group were intraperitoneal injected with cyclophosphamide 40,30,30 mg / kg,the blank control group was intraperitoneal injected with normal saline to prepare an animal model of qi-blood deficiency.Effect of An Shen Bu Nao oral liquid on anti-qi on mice model of qi and blood deficiencyDuring the administration period,the body weight of the mice was monitored,the behavioral state of the mice was observed,and the red blood cells(RBC)and the hemoglobin(Hg)content were measured every other day.On the 10 th day after the first administration,1 hour after the medicine,the eyeballs were taken and the blood was sacrificed.Rats,serum were separated,serum EPO,TPO,IL-2,IL-3 levels were determined,femurs were taken,bone marrow nucleated cells(BMC)were counted,thymus,liver and spleen were taken,and organ index was calculated.Effect of Anshen Bu Nao oral liquid anti-fatigue on mice model of qi and blood deficiencyThe body weight of the mice was monitored daily during the administration,and the behavioral state of the mice was observed.On the 10 th day after the first administration,the mice in each group were subjected to a rotating rod test and a weight-bearing swimming test,and the length of the rotating rod and the weight-bearing swimming time were recorded.At the end of the behavioral test,the liver and quadriceps of the mice were removed and the glycogen content in the liver and quadriceps was measured.Effect of Anshen Bu Nao oral liquid on sedation and tranquilization on mice model of qi and blood deficiencyThe body weight of the mice was monitored daily during the administration,and the behavioral state of the mice was observed.On the 10 th day after the first administration,the mice were subjected to autonomic activity test,pentobarbital sodium threshold dose hypnosis test,and sodium pentobarbital threshold.Under-dose hypnosis test,the number of mouse activities,sleep latency,sleep duration and the number of mice falling asleep were recorded.Results:1 Effect of ASBNY on anti qi-blood deficiencyThe mice in the control group had bright hair color,flexible activity,normal appetite and normal bowel movements.The mice in the model group were dull and easy to fall off,showing unpleasant to activity,get together,tired,arched,drowsiness,loss of appetite,and wet stools.Compared with the control group,the model group showed a significant decrease in body weight.Compared with the model group,An Shen Bu Nao A 5 m L/kg and B 10 m L/kg were increased the body weight(p<0.05),An Shen Bu Nao A 10 m L/kg and An Shen Bu Nao B 5 m L/kg had no effect on the body weight(p>0.05).Compared with the control group,the model group showed a significant decrease in red blood cell(RBC)and in hemoglobin(Hb)(p<0.01).Compared with the model group,An Shen Bu Nao A 10 m L/kg and B 10 m L/kg were significantly increased RBC and Hb(p<0.01);An Shen Bu Nao A 5 m L/kg and B 5 m L/kg had no effect on RBC in the mice of qi-blood deficiency(p>0.05).Compared with the control group,the model group showed a significant decrease in the number of bone marrow cells(BMC)(p<0.01).Compared with the model group,An Shen Bu Nao A 10 m L/kg and B 10 m L/kg were significantly increased the number of BMC(p<0.01).An Shen Bu Nao A 5 m L/kg and B 5 m L/kg had no effect on the number of BMC in the mice of qi-blood deficiency(p>0.05).Compared with the control group,erythropoietin(EPO),interleukin-3(IL-3)and a increase of interleukin-2(IL-2)were observed in the serum of Model group(p<0.05).Under the treatment with AnShenBuNao A 10 m L/kg and B 10 m L/kg,the level of EPO and IL-3 were significantly increased,IL-2 was increased in Model group(p<0.05 or p<0.01).Compared with the control group,the model group showed a decrease in thymus index and spleen index(p<0.05).An Shen Bu Nao A and B had no effect on thymus index and spleen index in the mice of qi-blood deficiency(p>0.05).2 Effect of ASBNY on anti-fatigue in qi-blood deficiency miceCompared with the control group,Model group mice needed the time on the rotated exhibited a significant reduction(p<0.01);An Shen Bu Nao A 10 m L/kg and An Shen Bu Nao B 5,10 m L/kg treatment increased the time on the rotated compared with Model group(p<0.05 or p<0.01).Compared with the control group,Model group mice needed more time to swim(p<0.05);An Shen Bu Nao A and B treatment increased the time on the swimming compared with Model group(p<0.05).Compared with the control group,a decrease of muscle glycogen were showed a decrease in model group(p<0.05),An Shen Bu Nao A 10 m L/kg and B 10 m L/kg treatment increased the level of muscle glycogen(p<0.05),An Shen Bu Nao A 5 m L/kg and B 5 m L/kg had no effect on muscle glycogen in the mice of qi-blood deficiency(p>0.05).Compared with the control group,a decrease of hepatic glycogen were showed a decrease in model group(p<0.01),An Shen Bu Nao A 10 m L/kg and B 10 m L/kg treatment increased the level of hepatic glycogen(p<0.05),An Shen Bu Nao A 5 m L/kg and B 5 m L/kg had no effect on hepatic glycogen in the mice of qi-blood deficiency(p>0.05).3 Effect of ASBNY on sedative in qi-blood deficiency miceCompared with the control group,the model group showed a significant decrease in the number of locomotor activity test(p<0.01).Compared with the model group,An Shen Bu Nao A 10 m L/kg and B 10 m L/kg were significantly increased the number locomotor activity test(p<0.01),An Shen Bu Nao A 5 m L/kg and B 5 m L/kg were increased the number locomotor activity test(p<0.05)Compared with the control group,the duration of sleep was showed a significant decrease in model group(p<0.01),An Shen Bu Nao A 10 m L/kg and B 10 m L/kg could increase the duration of sleep significantly(p<0.01).There was a tendency to prolong the duration of sleep,but there was no statistical significance in An Shen Bu Nao A 5 m L/kg and B 5 m L/kg(p>0.05).An Shen Bu Nao A and B had no effect on sleep latency in the mice of qi-blood deficiency(p>0.05).The number of animals falling asleep in control group was 2/10,in model group was 1/10.There were 4/10 and 7/10 in An Shen Bu Nao A 5 m L/kg and 10 m L/kg,there were 5/10 and 8 /10 in An Shen Bu Nao B 5 m L/kg and 10 m L/kg.Compared with the model group,An Shen Bu Nao A 10 m L/kg and B 10 m L/kg were significantly increased the number of animals falling asleep.(p<0.01).
