Objective:To understand the healthstatus of cadmium exposure population in Nandan,Hechi area.Furthermore,to detect the methylation level of CpG island of Klotho gene in the population,and to explore the effects of cadmium exposure on the health of the population and the methylation level of Klotho gene,and to provides a theoretical basis for further exploring the mechanism of cadmium exposure affecting the methylation of Klotho gene.Method:According to the difference of cadmium environmental pollution among different regions,the general population of Luoma,Chehe and Pingcun in Nandan county was selected as the research object.Perform a health checkup(including blood routine examination,liver function test and kidney function test,etc),and conduct questionnaire survey(including general demographic data and living environment information,etc.).DNA extracted from blood was used for pyrosequencing to detect the methylation level of CpG island of Klotho gene.And the concentration of cadmium in urine of subject was determined by atomic absorption spectrometry.Results: 1.There was a statistically significant difference in urinary cadmium levels in the three regions(P<0.001),followed by Luoma,Chehe and Pingcun from low to high.There were statistically significant differences in hemoglobin concentration,neutrophil count,monocytes count,platelet count,total protein and albumin concentrations in different cadmium exposed areas(P value <0.05).2.Cadmium exposure levels was positively correlated with mononuclear cell count(P = 0.001),and Cadmium exposure was negatively correlated with hemoglobin concentration,lymphocyte count,neutrophil count(P < 0.05).Cadmium exposure was positively correlated with urine ?2-microglobulin(P = 0.008),and negatively correlated with albumin(P = 0.03).There was a linear correlation between cadmium and hemoglobin concentration,lymphocyte,albumin count and platelet count(P <0.05).3.There was a statistically significant difference in the methylation level of Klotho gene in the three regions of Luoma,Chehe and Pingcun except pos.5 loci(all P values were <0.001).Cadmium exposure was positively correlated with the methylation rates of pos.2 and pos.6 locus(both P <0.05)and had a linear relationship(both P <0.05).The methylation levels of pos.2 and pos.4 sites of the Klotho gene were positively correlated with urine ?2-microglobulin(all P values were <0.05).Conclusion:There may be some relationship between cadmium exposure and abnormal blood system and liver and kidney function in the population in nandan area,and the hypermethylation of Klotho gene related to cadmium exposure may play a certain role.
