Objective:To investigate the expression level and cytobiological function of thrombospondin2(THBS2)in human cervical squamous cell carcinoma(SCC)SiHa cells. Methods: NC and THBS2 lentiviral vectors were constructed to infect SiHa cells respectively(named: NC-LV and THBS2-LV).The expression level of THBS2 were compared in SiHa cells before and after infection of NC-LV and THBS2-LV by using Western blot and Real time fluorescence quantitative PCR.CCK-8 ?transwell cell and scratch test were used to compare the proliferation ?invasion and migration ability of SiHa cells before and after infection of NC-LV and THBS2-LV.Flow cytometry was used to detect SiHa cell cycle and apoptosis of cervical squamous cell carcinoma before and after infection of NC-LV and THBS2-LV.Results: After infection with THBS2-LV,the expression of THBS2 in SiHa cells increased. After infection with THBS2-LV,the proliferation, invasion and migration ability of SiHa cells in cervical squamous cell carcinoma decreased. The apoptosis of SiHa cells in cervical squamous cell carcinoma increased after infection with THBS2-LV.Conclusion: THBS2 inhibits the proliferation, invasion and migration of SiHa cells in cervical squamous cell carcinoma.THBS2 may be a tumor suppressor gene of cervical squamous cell carcinoma and may be a potential therapeutic target for this disease.
