Study On The Relationship Between The Genetic Polymorphisms Of ESR1 Gene And The Susceptibility To Hepatocellular Carcinoma In The Population Of Fusui District Of Guangxi Province

Objective To investigate the relationship between single nucleotide polymorphisms(SNPs) of gene ESR1 and primary liver cancer developm- ent in the PLC group,the high risk group,normal group and the group had a family history of liver cancer of Fuisui District of Guangxi Province.Methads We studied 50 cases of PLC from Zhuang nationality , 50 cases of PLC from Han nationality, 24 cases of high-risk population from Zhuang nationality, 30 cases of normal population from Zhuang national- ity, 30 cases of normal population from Han nationality and 74 cases had a family history of liver cancer from Zhuang nationality. SNP locus rs9340772 and rs9322354 of ESR1 gene were selected as genetic markers.The polymorphism and mutational frequency of ESR1 gene were detected by the polymerase chain reaction combined with restriction fragment polymorphism(PCR-RFLP).Results (1)SNP locus rs9340772 of ESR1 gene in the PLC group,the high risk group,normal group and the group had a family history of liver cancer weren't mutation(.2)Among the PLC group,the high risk group,normal group and the group had a family history of liver cancer,the genetic mutation of SNP locus rs9322354 of ESR1 were 69%(69/100), 54.2%(13/24),58.3%(35/60) and 62.2%(46/74) respectively.There were no significant difference among them(χ2=2.933, P=0.402).(3)Among the PLC group of Zhuang nationality and Han nationality, the genetic mutation of SNP locus rs9322354 of ESR1 were 70%(35/50) and 68%(34/50) respectively , there were no significant difference (χ2=0.047, P=0.829).(4)Among the PLC group for HBsAg positive and HBsAg negative,the genetic mutation of SNP locus rs9322354 of ESR1 were 67.8%(57/84) and 75%(12/16) respectively , there were no significant difference (χ2=0.321, P=0.571).(5) Among the high risk group for HBsAg positive and HBsAg negative people had a family history of liver cancer,the genetic mutation of SNP locus rs9322354 of ESR1 were 54.2%(13/24) and 66%(33/50) respectively , there were no significant difference (χ2=0.965, P=0.326).(6) Among the normal group of Zhuang nationality and Han nationality, the genetic mutation of SNP locus rs9322354 of ESR1 were 66.7%(20/30) and 50.0%(15/30) respectively, there were no significant difference (χ2=1.714, P=0.8190).Conclusions (1)The SNP rs9340772 in ESR1 gene locus mutation did not happen in the normal population, the patients with liver cancer,high-risk population and the group had a family history of liver cancer of Zhuang and Han nationalities.It was indicated that the ESR1 gene SNP rs9340772 polymorphism was not related to the occurrence of primary liver cancer. (2)There were no significant difference in the SNP rs9322354 in ESR1 gene locus mutation among the normal population, the patients with liver cancer, high-risk population and the group had a family history of liver cancer of Zhuang and Han nationalities,so that, the SNP rs9322354 in ESR1 gene locus mutation was not related to the development of primary liver cancer. (3)The rate of gene mutation in ESR1 rs9322354 were no significant difference in the people with or without HBV infection whatever the patients of Zhuang and Han nationalities with liver cancer, high-risk groups or the people had a case history suffered from liver cancer. It was indicated that the hepatocarcin- ogenesis of HBV was not passed throught- out the mutation of rs9322354 in ESR1 gene,even though HBV infection is one pathogen of liver cancer development.
