Objective: To investigate liver kinase B1(LKB1)and stem cell markers[octamer-binding protein-4(OCT-4),NANOG,and Recombinant Sex Determining Region Y Box Protein 2(Resinant Sex Determining Region Y Box Protein 2,Expression of SOX2),Ki67] in Hepatocellular Carcinoma(HCC)Hepatocellular Carcinoma and Its Correlation with Prognosis in Patients with HCC.Methods: A total of 39 pairs of HCC patients with HCC who underwent hepatectomy from January 2013 to February 2013 in the Department of Hepatobiliary Surgery,Affiliated Tumor Hospital of Guangxi Medical University,and their corresponding para-cancerous tissue were collected and divided into liver cancer.Tissue group and paracancerous tissue group.Immunohistochemistry was used to detect the expression of LKB1,OCT-4,NANOG,SOX2,and Ki67 proteins in the two groups and their relationship with the prognosis of HCC patients was analyzed by Cox regression model.Kaplan-Meier method was used to calculate the relationship between LKB1 protein and survival rate of HCC patients.Spearman rank correlation analysiswas used to evaluate the correlation between LKB1 protein and OCT-4,NANOG,SOX2,Ki67 proteins in liver cancer tissues.Results: LKB1,OCT-4,NANOG,SOX2 and Ki67 proteins were expressed in liver cancer tissues.(1)Expression of LKB1 in both groups: LKB1 is simultaneously expressed in cytoplasm and nucleus.The positive expression rate of LKB1 in liver cancer tissues was 35.9%,and the positive expression rate in adjacent tissues was 79.5%(P<0.05).Univariate and multivariate analysis found that LKB1 was an independent protective factor affecting the prognosis of patients with HCC(P<0.05).Kaplan-Meier survival curve showed that the overall survival rate of patients with high LKB1 expression of HCC was better than that of patients with low expression of LKB1.Rate(P=0.049).(2)Expression of OCT-4 in two groups: OCT-4 is mainly expressed in the cytoplasm.The positive expression rate of OCT-4 in liver cancer tissues was69.2%,and the expression rate in adjacent tissues was 30.8%(P<0.05).Multivariate analysis found that OCT-4 was an independent factor affecting the prognosis of patients with HCC(P<0.05).(3)Expression of NANOG in two groups: NANOG is mainly expressed in the nucleus.The positive expression rate of NANOG in liver cancer tissues was 61.5%,and the positive expression rate in adjacent tissues was 76.9%(P<0.05).Multivariate analysis found that NANOG was an independent factor affecting the prognosis of patients with HCC(P<0.05).(4)Expression of SOX2 in two groups: SOX2 is mainly expressed in the nucleus.The positive expression rate of SOX2 in liver cancer tissues was 69.2%,and the positive expression rate in adjacent tissues was30.8%(P<0.05).Multivariate analysis found that SOX2 was not an independent factor affecting the prognosis of patients with HCC(P>0.05).(5)Expression of Ki67 in the two groups: Ki67 is mainly expressed in the nucleus.The positiveexpression rate of Ki67 in liver cancer tissues was 66.7%,and the positive expression rate in adjacent tissues was 71.79%(P<0.05).Multivariate analysis revealed that Ki67 was a long-term independent influence factor in patients with HCC(P<0.05).Conclusions:(1)LKB1 is lowly expressed in HCC tissues and highly expressed in adjacent tissues,which is negatively correlated with long-term survival rate of HCC patients,and is an independent influencing factor for long-term prognosis of HCC patients.(2)OCT-4,NANOG,SOX2,and Ki67 are highly expressed in liver cancer tissues and low in adjacent tissues,and have no correlation with LKB1 expression.(3)OCT-4,NANOG,and Ki67 are independent influencing factors for long-term prognosis in patients with HCC,but SOX2 is not an independent factor affecting the long-term prognosis of patients with HCC.
