Research objectsTo detect the expression of micro RNA-129-5p(MiR-129-5p)and E2F7 m RNA and protein in cancer tissues and adjacent tissues of rectal adenocarcinoma(READ),and analyze MiR-129-5p and E2F7 in READ organization.Whether the expression in the middle is correlated and the relationship between MiR-129-5p and E2F7 expression and the general clinicopathological features and prognosis of READ.MaterialThe specimens from the Zhengzhou University Affiliated Tumor Hospital from September 2016 to December 2016 were treated with 48 specimens of READ patients after surgical resection and some of them corresponded to adjacent tissues(generally selected upper rectal margins of rectal specimens)specimens.In 20 cases,the pathological types confirmed by preoperative pathology were rectal adenocarcinoma,and the pathological and clinical data of all READ patients were complete.All cases were followed up for 5-27 months,with a median follow-up of 24 months.MethodsReal-time Quantitative Polymerase Chain Reaction(q PCR)was used to detect the expression of MiR-129-5p and E2F7 m RNA in 48 READ cancer tissues and 20adjacent tissues.Using tissue microarray technology,immunohistochemistry(IHC)method was used to detect the expression of E2F7 protein in READ cancer tissues and adjacent tissues.The relationship between the expression of MiR-129-5p gene and E2F7 protein and the general clinicopathological features of READ patients was analyzed by chi-square test.The correlation between MiR-129-5p gene expression level and E2F7 m RNA expression level was analyzed by Pearson test.Two independent samples were used to analyze the expression of MiR-129-5p in READ tissue and adjacent tissues.Survival analysis was performed by KM curve and Log-rank test to investigate the expression of MiR-129-5p and E2F7 proteins.The impact of prognosis in patients with READ.Results:The expression level of MiR-129-5p in cancer tissues of patients with READ was significantly lower than that of adjacent tissues.The average expression of MiR-129-5p in 48 cases was 0.3069 ± 0.08167,and MiR-129-in 20 cases of adjacent tissues.The average expression of 5p was 0.7391 ± 0.09217,and the difference was statistically significant(P<0.05).The expression level of E2F7 m RNA in cancer tissues was significantly higher than that in adjacent tissues.The average expression of E2F7 m RNA in the 48 cases of cancer tissues was 2.362 ± 0.1333,and the average expression of E2F7 m RNA in 20 cases of adjacent tissues was 0.7403 ± 0.1382.The expression level of E2F7 m RNA was significantly higher than that of adjacent tissues,and the difference was statistically significant(P<0.05).The positive rate of E2F7 protein in 20 cases of adjacent tissues was 35.0%(7/20),and that in cancer tissues was 64.6%(31/48).The E2F7 protein was significantly higher in cancer tissues than in adjacent tissues,and the difference was statistically significant.(?2=5.766P=0.02).Correlation test analysis showed that MiR-129-5p expression level was negatively correlated with E2F7 m RNA expression level in READ tissues.The analysis of the relationship between the general clinicopathological features of 48 READ patients and the expression level of MiR-129-5p showed thatMiR-129-5p expression level and READ patient age,gender,tumor primary site,size,regional lymph node metastasis There was no significant difference in the degree of differentiation and distant organ metastasis(P>0.05).In the low expression group of MiR-129-5p,the depth of tumor invasion T3+T4 was 62.1%(18/29),and the proportion was significantly higher.The high expression group was 10.5%(2/19),and the difference was statistically significant(?2=12.547 P=0.01).The TNM stage III+IV was 42.1%(8/19)in the MiR-129-5p high expression group,the TNM stage III+IV was 75.9%(22/29)in the low expression group,and the proportion of the low expression group III+IV patients.Significantly higher than the high expression group,and the difference was statistically significant(?2=5.581 P=0.032).In the E2F7 protein positive group,the proportion of T3+T4 was 51.6%(16/31),the proportion of T3+T4 in the E2F7 protein negative group was only 17.4%(3/17),and the proportion of T3+T4 in the E2F7 protein positive group was significantly higher.In the E2F7 protein negative group,the difference was statistically significant(?2=5.296P=0.031).E2F7 protein expression was not associated with age,gender,primary site,size,degree of differentiation,regional lymph node metastasis,and distant organ metastasis.The difference was not statistically significant(P>0.05).The 2-year cumulative DFS of MiR-129-5p low expression group was 73.9%,and the cumulative DFS of MiR-129-5p high expression group was 84.2% for 2 years.There was no significant difference between the two groups(P=0.5334).The 2-year cumulative DFS rate was 83.9% in the E2F7 protein positive group and 76.5% in the E2F7 protein negative group.The difference between the two groups was not statistically significant(P=0.7636).Conclusions:1.Compared with adjacent tissues,micro RNA-129-5p was down-regulated and E2F7 m RNA was highly expressed in rectal adenocarcinoma tissues,and their expression levels were negatively correlated in rectal adenocarcinoma tissues.2.The low expression of micro RNA-129-5p in rectal adenocarcinoma is related to the depth of tumor invasion and TNM stage.The positive expression of E2F7protein is related to the depth of tumor invasion.
