recurrence rate provides reference and theoretical basis for the clinical treatment and research of flt3-itd mutagenesis positive AML with hexamethylhexaneMethods: 1.Clinical study: 30 patients with acute myeloid leukemia from Shanxi Province children's hospital blood disease hospital wards,were randomly divided into sinomenium acutum element has a combined chemotherapy in the treatment group and chemotherapy alone in the control group,each group of 15 patients with two groups in age and gender have no obvious differences on the illness severity,comparable to the control group of chemotherapy(induced ease with DA plans,strengthening maintain HA scheme)and support therapy such as anti-infection white blood transfusion treatment group on the basis of chemotherapy,The patients chemotherapy,for effective and 2 cases were markedly effective follow-up experiment research:(1)the four methyl azo thiazole blue experiment(determined by MTT)determination of different concentrations observation showed that :(1)both the clinical symptoms and signs of the patients in the experimental group and the control group were effectively improved,and the treatment group was superior to the control group in alleviating clinical symptoms and alleviating toxic and side effects of chemotherapy.The incidence of adverse reactionsin the treatment group was significantly lower than that in the control group,with significant differences between the two groups.(2)in the comparison of blood picture levels between the two groups before and after treatment,the recovery of hematopoietic function in the treatment group was significantly better than that in the control group after chemotherapy,and there was a significant difference between the two groups before and after treatment.(3)according to the qlq-c30 quality of life survey score table,compared with the overall quality of life improvement of patients in the two groups,the quality of life improvement of the treatment group was better than that of the control group,and there was a difference between the two groups before and after treatment.(4)in the treatment group,there were 12 cases of complete remission and 2 cases of partial remission,with a total effective rate of 93.34%;in the control group,there were 10 cases of complete remission and 3 cases of partial remission,with a total effective rate of 86.67%.The curative effect of the treatment group was significantly higher than that of the control group.The experimental results showed that:(1)MTT assay showed that Tet has an obvious inhibitory effect on the proliferation of MV411 cells,and the Tet gradually increases with the increase of drug duration and concentration,showing a time-dose dependence.The 48-hour IC50 of Tet was selected,and the value was 15.91nmol/L.(2)the results of flow cytometry showed that the average early apoptosis rates at the 48-hour time points of 0,20 and 40nmol/L Tet were(3.30 0.20)%,(17.10 0.36)% and(35.67 0.61)%,respectively.The mean early apoptosis rates at 72 HRS at 0,20,and 40nmol/L Tet were(7.37 0.32)%,(49.33 0.40)%,and(68.92 0.11)%,respectively.The results showed that the apoptosis rate increased with the increase of concentration and timeConclusion:1.Combined chemotherapy with hanfexetine a can significantly improve the CR and PR of patients with acute myeloid leukemia,relieve the clinical symptoms of patients,and have certain advantages over the adverse reactions after chemotherapy,improve the quality of life of patients during chemotherapy,and contribute to the recovery of hematopoietic function of patients with chemotherapy.Other serious adverse reactions have not occurred in clinical observation,which further indicates that combination of tetrandrine and chemotherapy is an effective and safe treatment for acute myeloid leukemia.2.Tet has a time-dose-dependent inhibitory effect on MV411 cell proliferation.Tet can induce apoptosis of MV411 cells.
