Objective:According to the Yanghongshan Longbiping granules effective substances,choosing the appropriate solvent on the optimal extraction process optimization,In order to make the granules,we choose suitable auxiliary materials and wetting agents,and establish the Yanghongshan Longbiping granules quality standards;The corresponding model was established to evaluate its inhibitory effect on prostate hyperplasia.To provide scientific basis for the development of a new drug candidate with stable and controllable quality,definite efficacy and clear mechanism of action to inhibit prostate hyperplasia.Methods1.Pharmaceutical experiments on Yanghongshan Longbiping granulesThe preparation methods of Yanghongshan Longbiping Granules:The comprehensive score of total flavonoids content and paste extraction amount was used as the index,On the basis of single factor,the orthogonal experiment was used to investigate the effects of three factors,including water content,extraction time and extraction times,on the total flavonoid extraction amount and paste extraction amount,and to optimize the extraction process.Granule molding rate as indicators to investigate the kinds and dosage of auxiliary materials,the concentration of the wetting agent to optimize the Yanghongshan Longbiping Granules the best molding process;By thin layer chromatography?TLC?of Yanghongshan Longbiping granules in Pimpinellathellungiana,Puerarin,Lycium,Crataegus pinnatifida and Panax quinquefolium L for qualitative identification;Using the UV-vis spectrophotometer?UV?and high performance liquid chromatography?HPLC?method,Detection of the content of flavonoids and the content of puerarin in theYanghongshan Longbiping Granules2.The pharmacodynamics and mechanism of Yanghongshan Longbiping granulesSD rats were subcutaneously injected with testosterone propionate once a day for30 days to establish a prostatic hyperplasia model in castrated rats.After successful replication of the model,each group was given corresponding drugs once a day for30d.After the last dose,the subjects fasted and could not help drinking water for 12h and weighed themselves.The prostate was completely removed after the blood was taken from the abdominal aorta.The wet weight was recorded by electronic balance and calculated the prostate index.The morphological changes of prostate tissues were observed by HE staining.Serum T,DTH,SOD,MDA,PACP,and the concentration of NO in prostate tissue were detected according to the kit instructions.Using Western blot method to detect the influence of the Nrf2/cox-2 protein expression level in prostate tissue.Results:1.Pharmaceutical experiments on Yanghongshan Longbiping granulesThe best extraction technology of the Yanghongshan Longbiping granules is obtained by orthogonal experiment byadd 12 times the amount of water,extracting3 times,each time 1 hour.Through the investigation of granule auxiliary materials and wetting agents,the final determination of granule molding technology is,powder:dextrin 1:0.4 mix,add appropriate amount of 85%ethanol soft material to get the highest molding rate of granule.the Yanghongshan Longbiping granules quality control:thin layer chromatography,according to the results of Yanghongshan Longbiping granules with Pimpinellathellungiana,Puerarin,Crataegus pinnatifida and Panax quinquefolium L control medicinal material shown on the corresponding position of the same color spots,and the negative without interference;but the negative of Lycium has interference to its;the Yanghongshan Longbiping granules calculated on dry goods,the total flavonoids content shall not be less than 39.072mg·g-1;The content of puerarin(C21H20O9)should not be less than 13.64mg·g-1.2.The pharmacodynamics and mechanism of Yanghongshan Longbiping granulesCastrated rats were induced by testosterone propionate to replicate prostatic hyperplasia.The experimental results showed that,compared with the model group,the high and medium dose groups could significantly reduce the prostate index of rats?p<0.05 or p<0.01?.The serum levels of T,DHT,PACP and MDA in high,medium and low dose groups were significantly decreased?p<0.01?.And high,medium and low dose of Yanghongshan Longbiping granules group could significantly increase model of rat prostate NO concentration?p<0.01?,and the marked increase in the level of serum SOD in rats.Western blot results showed that Nrf2/cox-2 protein expression level was significantly up-regulated in the model group compared with the blank control group?p<0.01?.Compared with model group high-dose sheep red shan uroschesis-particle group of Nrf2/Cox-2 protein expression level significantly lowered?p<0.05 or p<0.01?.Conclusion:In this paper,through the orthogonal experiment,single factor screening sheep red shan uroschesis flat particles the best extraction technology and molding process,and established the sheep red shan uroschesis flat grain quality evaluation method,for sheep red shan uroschesis flat particles quality control provides scientific theory basis;At the same time,this study confirmed the sheep red shan uroschesis flat particles has obvious inhibitory effect to the hyperplasia of prostate,its mechanism may be related to the sheep red shan uroschesis flat particles to adjust the body's hormone disorder,enhance the antioxidant capacity of the animal model.
