Pharmaceutical And Pharmacodynamic Reasearches On Jieyushunxin Granules

Jieyushunxin Granules are traditional Chinese medicines containing herbs:Radix Bupleuri Chinensis,Radix Salviae Miltiorrhizae,Rhizoma Acori Tatarinowii,Radix Curcumae Wenyujin,Radix Palygalae,Rhizoma Coptidis,Os Draconis,Concha Ostreae,Lapis Chloriti,which have the effect of dispersing the depressed liver-energy,clearing heat and eliminating phlegm,and tranquilizing mind,and are beneficial for the treatment of the depressive disorders due to pathogenic fire derived from stagnation of liver-qi and phlegm-heat attacking the heart,with manifestation of deprementia,restlessness,tantrum,gas pain of chest and hypochondrium,palpitation,insomnia,morbid forgetfulness,dreaminess,festless sleep,lassitude,and so on.Under the guidance of TCM theory and according to the State Food and Drug Administration,the article carries out the pharmaceutical and pharmacodynamic research using modern science and technology.1.Study of the preparationIn the study of preparation technology,according to the effectiveness of prescription indications,clinical drug characteristics and the nature of the active ingredients of drugs,we design preparation process:(1)We use the percolation of alcohol to extract Radix Salviae Miltiorrhizae and do experiments according to L9(34)perpendicular sheet containing four factors-the comminution of herbs,the density of alcohol,the amount of alcohol and speed of percolation,the results show that the best conditions are percolating the powder of Radix Salviae Miltiorrhizae with 20 times of 75%alcohol and speed 5 mL-min-1·kg-1.(2)We choose trational water abstraction methods to extract Radix Bupleuri Chinensis,et al.and do experiments according to L9(34)perpendicular sheet containing three factors-the amount of water,abstraction time and abstraction frequency.The best water abstraction schema is extracting them with water of 10 times for 3 times,1.5 hours once.(3)We choose wet method the to make granula,and do experiments to preffer accessory and corrigent by forming,bulk density,angle of repose and melting hygroscopic.The best accessory is lactose,corrigent is stevia sugar.In order to test the rationality of the preparation,we have research on mid-experiment for three times,and the products are detected according to all the quality control.The the pratical production is feasible and the received products rate is above 90%.2.Study of the quality standard.We determined the index and methods of quality control.We use TLC toidentify Radix Salviae Miltiorrhizae,Rhizoma Acori Tatarinowii,Radix Curcumae Wenyujin,Radix Palygalae,Rhizoma Coptidis.et.al.The results show that the spots are clear on the TLC picture and the negative control was non-interference,so we could use it as the qualition identification.HPLC is used to assay Salvianolic acid B.The results show that the contents in every capsule of Salvianolic acid B shouldn't be less than 150.0mg and.It says the quality of this preparation is stable.We check the moisture and melting,et al of Jieyushunxin Granules at last,too.3.Study of the stabilityIn order to examine the stability of the medicine and determine the time of validity of the medicine,we keep the wrapping material at indoor temperature and detect three batches of products according to all the indexes from the quality control.4.Study of pharmacodynamicWe took the despair model in mouse,the reserpine antagonism model in mouse,the chronic stress model of depression mouse models to provide the basis to verify the efficacy of Jieyushunxin Granules.The results showed that the mechanism of antidepressant effect may be acting on the hypothalamus-pituitary-adrenal axis(HPA).
