Objective: To explore the regulatory role of micro RNA-365a-3p in the degeneration of lumbar facet cartilage,and to provide a potential molecular basis for the treatment of lumbar facet osteoarthritis.Methods: Twenty patients with Lumbar degenerative diseases and 10 patients with lumbar fracture were selected for surgical treatment.All patients were examined by CT and MRI before operation.The patients were divided into two groups according to the severity of degeneration(degeneration group and normal group),and the articular cartilage of lumbar facet was obtained and separated under aseptic condition.Tissue and specimen were stained with safranine and green fixing;differential expression of micro RNA-365a-3p in normal and degenerative cartilage tissue was observed by q RT-PCR;articular chondrocytes were cultured in vitro under aseptic conditions and treated with IL-1beta,respectively.Micro RNA-365a-3p mic and inhibitor were transfected into chondrocytes in vitro,and chondrocyte proliferation was detected by CCK8 method;q RT-PCR and Western blot were used to detect the expression of micro RNA-365a-3p in normal and degenerative cartilage tissue.The differential expressions of collagen II,proteoglycan and MMP-13 were detected before and after overexpression and overinhibition.Results: The lumbar facet cartilage tissues were divided into two groups by CT,MRI and pathological staining.RT-PCR showed that the expression of micro RNA-365a-3p was significantly down-regulated in degenerated facet articular cartilage tissues,and differentially expressed.Normal facet articular chondrocytes were obtained in vitro under sterile conditions.After different treatments with IL-1beta,micro RNA-365a-3p was overexpressed and inhibited.-At 365a-3p,the proliferation rate of chondrocyte was significantly increased in the mimic group.Western blot and q RT-PCR showed that the expression of collagen II and proteoglycan in the mimimic overexpression group was significantly increased,while the expression of matrix metalloproteinase-13(MMP-13)was significantly down-regulated.Conclusion: The expression of micro RNA-365a-3p in cartilage tissue of patients with severe degeneration of lumbar facet joints decreased significantly,and micro RNA-365a-3p could increase the proliferation rate of articular facet chondrocytes and enhance the synthesis of cartilage matrix.
